چکیده

The periopathogen Aggregatibacter actinomycetemcomitans is associated with aggressive periodontitis, a severe disease that causes rapid tooth loss among young adolescents. A. actinomycetemcomitans produce and secrete leukotoxin a pore forming protein which causes death of human polymorphonuclear leukocytes (PMNs) such as neutrophils. PMNs are the most abundant leukocyte and have important defence mechanisms such as formation of neutrophil extracellular traps (NETs) used to trap pathogens and the release of chemokines, such as IL-8. The aim of the study was to investigate the host-pathogen interaction between PMNs and A. actinomycetemcomitans focusing on the role of leukotoxin-induced NET-formation under anaerobic conditions. This study has examined the impact of high-leukotoxic JP2-clones and low-leukotoxic non-JP2 clones of A. actinomycetemcomitans as well as purified leukotoxin. Based on previous studies performed under aerobic conditions, we hypothesized that the highleukotoxic clones would induce greater production of NETs than low-leukotoxic clones. A second hypothesis was that PMNs stimulated with purified leukotoxin would produce more NETs than unstimulated controls. PMNs from healthy donors were infected with A. actinomycetemcomitansstrains or stimulated with leukotoxin isolated from high-leukotoxic JP2-clones collected from patient samples. NETs were quantified using immunofluorescent microscopy and IL-8 release was measured using ELISA. Our results showed a trend of increased NET-formation during A. actinomycetemcomitans infection and an increased NET-formation upon leukotoxin stimulation under both aerobic and anaerobic conditions. Untreated PMNs released low basal levels of IL-8 while A. actinomycetemcomitans infected PMNs released higher levels under aerobic conditions, whereas under anaerobic conditions the results were more uncertain.

Neutrophil-derived resistin release induced by Aggregatibacter actinomycetemcomitans.

Resistin is an adipokine that induces insulin resistance in mice. In humans, resistin is not produced in adipocytes, but in various leukocytes instead, and it acts as a proinflammatory molecule. The present investigation demonstrated high levels of resistin in culture supernatants of neutrophils that are stimulated by a highly leukotoxic strain of Aggregatibacter actinomycetemcomitans. In contr...

Effects of Aggregatibacter actinomycetemcomitans leukotoxin on neutrophil migration and extracellular trap formation

BACKGROUND Aggressive periodontitis is associated with the presence of Aggregatibacter actinomycetemcomitans, a leukotoxin (Ltx)-producing periodontal pathogen. Ltx has the ability to lyse white blood cells including neutrophils. OBJECTIVES This study was aimed at investigating the interactions between neutrophils and Ltx with regard to the chemotactic properties of Ltx and the release of neu...

Aggregatibacter actinomycetemcomitans arcB influences hydrophobic properties, biofilm formation and adhesion to hydroxyapatite

The regulation of gene expression in the oral pathogen Aggregatibacter actinomycetemcomitans is still not fully elucidated. ArcAB is a two-component system which allows facultative anaerobic bacteria to sense various respiratory growth conditions and adapt their gene expression accordingly. This study investigated in A. actinomycetemcomitans the role of ArcB on the regulation of biofilm formatio...

Nucleases from Prevotella intermedia can degrade neutrophil extracellular traps

Periodontitis is an inflammatory disease caused by periodontal bacteria in subgingival plaque. These bacteria are able to colonize the periodontal region by evading the host immune response. Neutrophils, the host's first line of defense against infection, use various strategies to kill invading pathogens, including neutrophil extracellular traps (NETs). These are extracellular net-like fibers c...
