چکیده

Methionine was found to be more efficient than formate, serine, bicarbonate, formaldehyde, or choline as a source of the C-28 methyl group of ergosterol in yeast (1). While transfer of an intact methyl group to carbon has not been previously recorded, oxidation of the methyl group of methionine to formate has been demonstrated and formate-Cl* has been shown to give rise to C-28 of ergosterol (2). Nevertheless, preliminary experiments in this laboratory indicated that a true transmethylation may occur (3). The experiments described in this paper were designed to elucidate the mechanism of the transfer of the l-carbon unit from methionine to sterol.

approved Major Professor An enzyme system from cell -free yeast homogenates mediating the transfer of the methyl group from Sadenosylmethioninemethyl -C14 or methionine -methyl -C14 to C -28 of ergosterol has been studied. The effects of divalent cations, suithydryl inhibitors and reducing substances on the activity of the enzyme system have been investigated. It was established that for maxima...

Transmethylation of sterols in aerobically adapting Saccharomyces cerevisiae.

The transmethylation of methyl-(14)C-methionine and methyl-(14)C-adenosylmethionine into the nonsaponifiable lipids of anaerobically grown yeast during adaptation to aerobic conditions was investigated. The rate and extent of methyl transfer increased with aeration time and was dependent upon the presence of a fermentable carbon source and O(2). Methionine and adenosylmethionine uptake rates in...

Inhibition of sterol transmethylation by S-adenosylhomocysteine analogs.

Structural analogs of S-adenosylhomocysteine were tested in vitro for inhibition of the yeast S-adenosylmethionine:delta 24-sterol-C-methyltransferase enzyme. A wide inhibitory range by these compounds was observed, suggesting which structural features of the parent compound are important for binding to the enzyme. No analog tested had inhibitory activity specific only for this enzyme. The most...

Application of sterol synthesis inhibitors to investigate the sterol requirements of protozoa and plants.

T h e major sterols found in higher plants. algae. fungi and many protoma are typically substituted at C-24 in the sidechain by a methyl or ethyl group. Depending upon the organism, the C-24 alkyl sterol can have either the 2311or 24s-configuration and the extra carbons are derived by transmethylation reactions involving S-adenosylmethionine and appropriate A'4and A''''x -sterol substrates 11 -...

A new triazole, voriconazole (UK-109,496), blocks sterol biosynthesis in Candida albicans and Candida krusei.

Voriconazole (UK-109,496) is a novel triazole derivative with potent broad-spectrum activity against various fungi, including some that are inherently resistant to fluconazole, such as Candida krusei. In this study we compared the effect of subinhibitory concentrations of voriconazole and fluconazole on sterol biosynthesis of fluconazole-resistant and -susceptible Candida albicans strains, as w...
