Cell-free Translation of Cricket Paralysis Virus RNA: Analysis of the Synthesis and Processing of Virus-specified Proteins
نویسندگان
چکیده
Translation of cricket paralysis virus (CrPV) RNA in a rabbit reticulocyte lysate yielded proteins which co-migrate on polyacrylamide gels with the structural proteins VP 1 and VP3. These proteins were identified by partial proteolysis and a number of high mol. wt, proteins were shown to be precursors of the structural proteins. The third structural protein VP2 was not identified in vitro but was shown to be formed in vivo from the minor 'structural' protein VP0. The profile of high mol. wt. proteins in vitro differed from those found in infected cells, but processing was similar with precursors being cleaved sequentially to give a group of proteins with mol. wt. of 50000 to 63 000. Processing proceeded in vitro to give VP1 and VP3 but synthesis of VP2 and its immediate precursor VP0 was not apparent. This is consistent with an inhibition of synthesis of VP0 found in infected cells treated with iodoacetamide and suggests that CrPV utilizes a different mechanism for the synthesis of VP0 and VP2 than it does for VP1 and VP3. A tentative model for the processing of CrPV-specified proteins is proposed.

Construction of a Minigenome Rescue System for Measles Virus, AIK-c Strain
Background:In the recent decade, the reverse genetics method has been broadly used for rescue of negative-stranded RNA viruses from cDNA or viral minigenomes. This technique has been applied to study different steps in virus replication and virus-host interactions. Reverse genetics could also be implemented for design of new vaccines. The T7 RNA polymerase activity as well as virus (nucleocapsi...
متن کاملIn vitro Expression Studies of Three Proteins of Iranian Wheat Stripe Virus
The genome of Iranian wheat stripe virus (IWSV), a tentative member of the genus Tenuivirus, is comprised of three ambisense and one negative sense RNA segments. The coat and non-structural proteins encoded by the vcRNA3 and vRNA4 genes, respectively, were efficiently translated in vitro. Translated proteins of vcRNA3 and vRNA4 transcripts were approximately 35000 and 22000 in Mr, respectively,...
متن کاملTemporal Regulation of Distinct Internal Ribosome Entry Sites of the Dicistroviridae Cricket Paralysis Virus.
Internal ribosome entry is a key mechanism for viral protein synthesis in a subset of RNA viruses. Cricket paralysis virus (CrPV), a member of Dicistroviridae, has a positive-sense single strand RNA genome that contains two internal ribosome entry sites (IRES), a 5'untranslated region (5'UTR) and intergenic region (IGR) IRES, that direct translation of open reading frames (ORF) encoding the vir...
متن کاملThe IRES5′UTR of the dicistrovirus cricket paralysis virus is a type III IRES containing an essential pseudoknot structure
Cricket paralysis virus (CrPV) is a dicistrovirus. Its positive-sense single-stranded RNA genome contains two internal ribosomal entry sites (IRESs). The 5' untranslated region (5'UTR) IRES5'UTR mediates translation of non-structural proteins encoded by ORF1 whereas the well-known intergenic region (IGR) IRESIGR is required for translation of structural proteins from open reading frame 2 in the...
متن کاملMolecular detection of Acute Bee Paralysis Virus in Iran
Acute bee paralysis virus (ABPV) is a small single stranded RNA virus recently classified within the family Dicistroviridae, genus Cripavirus. Here, we describe the first study of ABPV in unhealthy bee colonies, which has been an unusual loss in adult bee population and significant honey bee mortality during the year. The aim of this study was evaluation of ABPV infection in honey bee colonies ...
