Leukemia: Serial Transplantation of Human Leukemic Lymphoblasts in the Newborn Syrian Hamster1

چکیده
Human leukemia cells are being regularly grown intraperitoneally in the intact newborn Syrian hamster as 3 serially transplantable sublines of lymphosarcoma. These sublines, established from suspension cultures of human leukemic lymphoblasts (CCRF-CEM cells) by the implantation of large numbers of cells, exhibit similar transplantation and growth characteristics, and are presently in their llth, 8th, and 6th serial passages, respec tively. The intraabdominal lymphosarcomas frequently metastasize to the central nervous system, and in some instances progress to leukemia—a striking recapitulation of the human disease from which the CCRF-CEM cells were originally isolated. Immunofluorescence evidence clearly indicates the presence of human species-specific antigens in the cells of the serially trans planted lymphosarcomas. The morphology of these cells, as determined by electron micrography, does not differ significantly from that of the CCRF-CEM cells in suspension culture. There is as yet no morphologic evidence for the presence of virus asso ciated with growth of these cells, either in suspension cultures or

متن کاملThe effect of resveratrol on the expression of MDR1 gene in leukemic lymphoblast’s of acute lymphoblastic leukemia patients
Background: Chemotherapy plays a very important role in the treatment of leukemia but the resistance properties of the lymphoblasts limit the effect of chemotherapy. One of the main mechanisms of resistance to chemotherapy is the increased expression of MDR1 gene. The aim of this study was to explore the effect of resveratrol on the expression of MDR1 gene in leukemic lymphoblast of new...
متن کاملHeterotransplantation of spontaneous leukemia of AKR mice into newborn rats.
Spontaneous lymphoid leukemia of the inbred AKR strain was transplanted into newborn rats, not later than 24 hours after birth. Two intraperitoneal or intravenous injections, spaced 7-10 days apart, resulted in progressive, fatal growth. Single subcutaneous or intraperitoneal injections of leukemic cell suspensions did not establish permanent heterografts. Serial passage was successful in newbo...

Lymphoblasts obtained directly from the peripheral blood buffy coat of a child with acute lymphoblastic leukemia partially or completely repopulated the depleted bone marrow spaces of 2 hamsters i.v. infused shortly after whole body irradiation in the supralethal dose range. A more extensive series of experiments with lymphoblasts (CCRF-CEM) isolated directly in continuous suspension cultures f...
متن کاملEpigenetic effects of decitabine on acute lymphoblastic and acute promyelocytic leukemia cells
Background: Decitabine (5-aza-2'-deoxycytidine, DAC) is a deoxycytidine analog currently used as an effective drug against myelodysplastic syndromes and acute myeloid leukemia. Although various studies have pointed out the epigenetic effects of this drug, its epigenetic mechanisms in different leukemic cell lines are not specified. In this lab trial study, possible epigenetic effects of decitab...
