چکیده

What are ectosomes? Ectosomes are vesicles of various size (0.1–1mm in diameter) that bud directly from the plasma membrane and are shed to the extracellular space. At variance with living cells, ectosomes have on their surface the phospholipid phosphatidylserine. These vesicles were long considered to be artefacts, and then they were confused with exosomes — the vesicles discharged upon exocytosis of multivesicular bodies — and with cytoplasmic particles generated during apoptosis. Now, ectosomes attract great interest: of the hundreds of papers published so far on ectosomes, over 50% have appeared in just the last two years.

Ectosomes released by human neutrophils are specialized functional units.

Here we show that human polymorphonuclear leukocytes (PMN) release ectosomes independently of complement attack during their activation both in vitro and at the site of inflammation in vivo. Patterns of biotinylated proteins on the surface of PMN and on PMN-derived ectosomes indicated a specific sorting of cell surface proteins into and out of ectosomes. Ectosomes expressed clusters of compleme...

Activated polymorphonuclear neutrophils disseminate anti-inflammatory microparticles by ectocytosis.

On activation, human neutrophils release microparticles, called ectosomes, directly from the cell surface membrane. Microparticles from platelets, endothelial cells, and monocytes were reported to support coagulation or to modulate vascular homeostasis by activating monocytes as well as endothelial cells. We find that neutrophil ectosomes have no proinflammatory activity on human macrophages as...

Proteogenomic analysis reveals exosomes are more oncogenic than ectosomes

Extracellular vesicles (EVs) include the exosomes (30-100 nm) that are produced through the endocytic pathway via the multivesicular bodies and the ectosomes (100-1000 nm) that are released through the budding of the plasma membrane. Despite the differences in the mode of biogenesis and size, reliable markers that can distinguish between exosomes and ectosomes are non-existent. Moreover, the pr...

Mycobacterium tuberculosis-induced neutrophil ectosomes decrease macrophage activation.

BACKGROUND The existence of ectosome-like microvesicles released by neutrophils was proposed a few decades ago. Other studies revealed that the innate immune response during mycobacterial infection is accompanied by an intense migration of neutrophils to the site of infection, which may be important during the acute phase of tuberculosis. We found that the ectosomes derived from infected neutro...

Ectosomes and exosomes: shedding the confusion between extracellular vesicles.

Long- and short-distance communication can take multiple forms. Among them are exosomes and ectosomes, extracellular vesicles (EVs) released from the cell to deliver signals to target cells. While most of our understanding of how these vesicles are assembled and work comes from mechanistic studies performed on exosomes, recent studies have begun to shift their focus to ectosomes. Unlike exosome...
