RNAi2010: Gene Regulation by Small RNAs
نویسندگان
چکیده
The Argonaute proteins constitute a highly conserved family of nucleic acid-binding proteins whose members have been implicated in RNA interference (RNAi) and related phenomena in several organisms. In particular, Argonaute 2 (Ago2) represents one of the key players in the RNA induced silencing complex (RISC). However, recently published literature describes that Ago2 itself is regulated under certain cellular conditions by post-translational modifications. In this work, we investigated the activity of human Ago2 under various conditions of cell stress. Under such conditions, the sub-cellular localization of Ago2 was altered which was coincident with its decreased function in the RNAi pathway. This work implies that specific cellular stress conditions induce an intracellular translocation of Ago2 protein excluding it from sites of action of the RNAi machinery. Details will be presented and discussed. J RNAi Gene Silencing (2010), 6(1), 367 Workflow for identifying miRNA function and targets in EMT

متن کاملEvaluation of the role of mico-RNAs in cardiomyocytes differentiation of mesenchymal stem cells
Stem cells are a good alternative for regenerative medicine because of their characteristics such as self-renewal and differentiation potential. They are classified into different types of stem cells including embryonic stem cells, induced pluripotent stem cells, multipotent stem cells, and ultimately uni-potent stem cells. Mesenchymal stem cells extracted from adult tissues. Due to the lack of...
متن کاملP-44: Investigation The Role of MicroR-NAs in Spermatogenesis and Male Infertility
Background: One of the most remarkable processes during spermatogenesis is the drastic change in gene expression. It has been recently proposed that small RNAs such as micorRNAs (miRNAs) contribute a great deal of function in regulation of the gene expression in sperm. Alterations in miRNAs function prove to be detrimental to proper spermatogenesis. As many patients suffer from idiopathic infer...
متن کاملSmall interfering RNA; principles, applications and challenges--
Gene silencing using RNAi (RNA interference), has recently been used as a successful laboratory technique in determining the function and control of gene expression and provides a wide range of applications in molecular biology and gene therapy. RNAi is a method of suppressing gene expression. In this direction, a single-stranded RNA molecule of about 21–23 nucleotides, called siRNA (small inte...

Background: Dendritic cells (DCs) are ideal accessory cells in the field of gene therapy. Delivery of DNA and siRNA into mammalian cells is a useful technique in treating various diseases caused by single gene defects. Selective gene silencing by small interfering RNAs (siRNAs) and antisense oligodeoxynucleotides (ODN)s is an efficient method for the manipulation of cellular functions. An effic...
متن کامل[Advances in mechanism of small RNAs].
The discovery of RNA interference (RNAi) heralded a revolution in RNA biology. Researchers uncovered 'hidden' layers of regulation of gene expression, in which many previously unidentified families of small RNAs (consisting of approximately 20-30 nucleotides) mediate gene silencing in transcriptional and post-transcriptional levels. In eukaryotes, these small RNAs, including siRNAs, miRNAs, piR...
