Heterogeneous Claudin-1 Expression in Human Liver
نویسندگان
چکیده
human liver. Representative images of claudin-1 costained with markers specific for apical (CD10) or basolateral (cytokeratin 8) membranes. Heterogeneous patterns of claudin-1 at apical and basolateral membranes were observed. Magnified volumetric images (white box) prepared using Imaris software demonstrate claudin-1 localization around CD10, in keeping with a pericanalicular tight junction distribution. Application of low and high fluorescent intensity thresholds discriminates apical tight junction-associated (silver) and basolateral (yellow) pools of claudin-1. Nuclei were stained with DAPI (blue) and the scale bar represents 10 lm. Heterogeneous Claudin-1 Expression in Human Liver

Comparison of the Expression of Hepatic Genes by Human Wharton's Jelly Mesenchymal Stem Cells Cultured in 2D and 3D Collagen Culture Systems
Background: Human Wharton’s jelly mesenchymal stem cells (HWJMSCs) express liver-specific markers such as albumin, alpha-fetoprotein, cytokeratin-19, cytokeratin-18, and glucose-6-phosphatase. Therefore, they can be considered as a good source for cell replacement therapy for liver diseases. This study aimed to evaluate the effects of various culture systems on the hepatocyte-specific gene expr...
متن کاملSex-Dependent Claudin-1 Expression in the Liver of Euthyroid and Hypothyroid Mice.
BACKGROUND In the liver the tight junction protein claudin-1 plays an important role in bile secretion by maintaining the paracellular barrier of bile canaliculi and the bile duct. A diminished bile excretion has been found in hypothyroid patients, and the prevalence of gallstones is increased in hypothyroidism. This association, however, only applies for men and is in contrast to the well-esta...
متن کاملTranscriptional activation of the human claudin-18 gene promoter through two AP-1 motifs in PMA-stimulated MKN45 gastric cancer cells.
Claudin-18 (CLDN18), a member of the claudin family of proteins that are structural components of tight junctions, has two alternatively spliced variants, claudin-18a1 and claudin-18a2, which are highly expressed in lung and stomach, respectively. Downregulation of claudin-18a2 is associated with gastric cancers of an intestinal phenotype; however, the mechanisms regulating its expression have ...
متن کاملReduced expression of the claudin-7 gene correlates with venous invasion and liver metastasis in colorectal cancer.
Claudins, members of a large family of adherent junction proteins, regulate the integrity and function of tight junctions and influence tumorigenesis. Studies have suggested that altered levels of different claudins are related to carcinoma-cell invasion and disease progression. This study examined the relationship between the relative expression of claudin genes and clinicopathological factors...
متن کاملExpression of claudins-1, -4, -5, -7 and occludin in hepatocellular carcinoma and their relation with classic clinicopathological features and patients' survival.
BACKGROUND Occludin and claudins are integral constituents of tight junction proteins and are de-regulated in various malignancies, including hepatocellular carcinoma (HCC). This study investigated whether expression of claudins 1, 4, 5, 7 and occludin may be used as prognostic markers for overall and disease-free survival in patients with HCC after hepatectomy. PATIENTS AND METHODS The study...
