Review: cholinesterase inhibitors have a modest effect on neuropsychiatric and functional outcomes in Alzheimer's disease.

چکیده
Data extraction Two reviewers extracted data independently about study methods, sources of bias, and outcomes. Sixteen trials included neuropsychiatric outcomes and 18 included functional measures. Neuropsychiatric outcomes were measured with the Neuropsychiatric Inventory (NPI) and the Alzheimer Disease Assessment Scale, noncognitive (ADAS-noncog). Data were combined using weighted mean differences. Functional outcomes were measured with activities of daily living and instrumental activities of daily living scales. Data were analysed using standardised mean differences.

Efficacy of cholinesterase inhibitors in the treatment of neuropsychiatric symptoms and functional impairment in Alzheimer disease: a meta-analysis.
CONTEXT Cholinesterase inhibitors are the primary treatment for the cognitive symptoms of Alzheimer disease (AD). Cholinergic dysfunction is also associated with neuropsychiatric and functional deficits, but results from randomized controlled trials of cholinesterase inhibitors are conflicting. OBJECTIVE To conduct a systematic review and meta-analysis to quantify the efficacy of cholinestera...

Background We investigated the effect of cholinesterase inhibitors on all-cause discontinuation, efficacy and safety, and the effects of study design-, intervention-, and patient-related covariates on the risk-benefit of cholinesterase inhibitors for Alzheimer's disease. Methods A systematic review and meta-analysis of randomized placebo-controlled clinical trials comparing cholinesterase inh...
متن کاملThe recent development in synthesis and pharmacological evaluation of small molecule to treat Alzheimer's diseases: A review
Alzheimer's disease is a neurological disorder in which the death of brain cells causes memory loss and cognitive decline. A neurodegenerative type of dementia, the disease starts mild and gets progressively worse. Like all types of dementia, Alzheimer's is caused by brain cell death. The most common presentation marking Alzheimer's dementia is where symptoms of memory loss are the most promine...

متن کاملProgress update: Pharmacological treatment of Alzheimer’s disease
A number of drugs have been approved for the treatment of Alzheimer's disease (AD) and a larger number are being studied as possible therapies. The current mainstays of the pharmacotherapy of AD are the cholinesterase inhibitors (donepezil, galantamine, rivastigmine) and memantine. They collectively have acceptable tolerability and proven but modest efficacy. The agents being studied include di...
