Brilliant Cresyl Blue in Sap of Nitella

چکیده
It has been shown by several investigators I that when the pl-I value of the external dye solutions is increased, the rate of accumulation of a basic dye in the cell sap is accelerated. Two different explanations for this have been given. Some have accounted for it on the basis that the basic dye enters the cell in the form of a dye hydrate and combines with the substances in the cell; and that, therefore, the increase in the rate of accumulation of the dye is due to the increase in the concentration of the dye hydrate. Others ~ have assumed that the increase in the rate of accumulation of the dye is due primarily to the increase in the concentration of the combining substances in the cell caused by the increase in the pH value of the cell contents. From these standpoints it is of decided interest to see what will happen if we increase the pH value of the cell sap, while keeping tha t of the external solution constant. An investigation of this sort was made by McCutcheon and Lucke, 3 but from their

Accumulation of Brilliant Cresyl Blue in the Sap of Living Cells of Nitella in the Presence of Nh3
When the living cells of Nitella are placed in a solution of brilliant cresyl blue containing NH(4)Cl, the rate of accumulation of the dye in the sap is found to be lower than when the cells are placed in a solution of dye containing no NH(4)Cl and this may occur without any increase in the pH value of the cell sap. This decrease is found to be primarily due to the presence of NH(3) in the sap ...
متن کاملMechanism of the Accumulation of Dye in Nitella on the Basis of the Entrance of the Dye as Undissociated Molecules
The rate of penetration of brilliant cresyl blue into the living cells of Nitella indicates that the dye enters only in the form of the undissociated molecule. At equilibrium the total concentration of the dye in the sap is proportional to the concentration of the free base in the outside solution.
متن کاملA GLASS ELECTRODE APPARATUS FOR MEASURING THE pH VALUES OF VERY SMALL VOLUMES OF SOLUTION
A glass electrode apparatus is described with which pH measurements can be made with as small volumes as 2 drops (about 0.14 cc.) of solution. Using this apparatus the change of pH of the vacuolar sap of Nitella, due to the penetration of brilliant cresyl blue, has been readily followed. The sap and the dye have been found to poison the usual type of hydrogen electrode.
متن کاملStudies of the Inner and Outer Protoplasmic Surfaces of Large Plant Cells
The vacuolar surface of Nitella is covered with a non-aqueous film too thin to be visible as a separate membrane. The motion of the protoplasm may subject this film to a good deal of mechanical disturbance. Apparently this does not rupture the film for no dye escapes into the protoplasm as the result of such disturbance when the vacuolar sap is deeply stained with neutral red or brilliant cresy...
متن کاملOn the Accumulation of Dye in Nitella
Living cells of Nitella were placed in different concentrations of brilliant cresyl blue solutions at pH 6.9. It was found that the greater the concentration of the external dye solution, the greater was the speed of accumulation of the dye in the cell sap and higher was the concentration of dye found in the sap at equilibrium. Analysis of the time curves showed that the process may be regarded...
متن کاملThe Effect of Acetate Buffer Mixtures, Acetic Acid, and Sodium Acetate, on the Proto- Plasm, as Influencing the Rate of Penetration of Cresyl Blue into the Vacuole of Nitella. by Marian Irwin
An adequate theory of the penetration 1,~,3 of cresyl blue into the vacuole of living cells of the fresh water plant Nitella may be stated as follows. The dye, in form of free base, which predominates at a high pH value, diffuses in and out of the vacuole very freely, while the dye in form of salt, which predominates at a low pH value, diffuses so slowly that its rate of penetration and that of...
