The identification of monoconjugates of bilirubin in bile as amide derivatives.

چکیده
Recent reports have shown that bilirubin can be esterified in vivo to form a variety of acyl glycosides (Kuenzle, 1970; Heirwegh, Van Hees, Leroy, Van Roy & Jansen, 1970). The question as to whether these conjugates are present in bile as monoconjugates as well as diconjugates remains in doubt. Indirect evidence for the existence of bilirubin monoglucuronide, based on the estimation of the molar ratio of bilirubin to glucuronic acid and on the production of equal amounts of azobilirubin and of azobilirubin glucuronide on diazotization of an isolated bilirubin conjugate, has been presented by several groups of workers (Billing, Cole & Lathe, 1957; Giovanetti, Maggiore & Vivaldi, 1961; Jacob-

Unconjugated bilirubin and an increased proportion of bilirubin monoconjugates in the bile of patients with Gilbert's syndrome and Crigler-Najjar disease.
Bilirubin pigments were studied in the bile of 20 normal adults, 25 patients with Gilbert's syndrome, 9 children with Crigler-Najjar disease, and 6 patients with hemolysis, to determine how a deficiency of hepatic bilirubin UDP-glucuronosyltransferase would affect the end products of bilirubin biotransformation. In the bile from patients with Gilbert's syndrome, a striking increase was found in...
متن کاملBilirubin conjugates in bile of man, rat and dog. Semi-quantitative analysis of bile composition by thin-layer chromatography.
1. Conjugated bile pigments, separated in two fractions by semi-quantitative t.l.c. performed on silicic acid with phenol/water as the developing solvent, were treated with diazotized ethyl anthranilate. Resulting dipyrrylazo derivatives were analysed by quantitative t.l.c. 2. The tentative structure elucidation of tetrapyrrolic bilirubin conjugates and semi-quantitative evaluation of rat bile,...
متن کاملSeparation by thin-layer chromatography and structure elucidation of bilirubin conjugates isolated from dog bile.
1. A system for separation of bile pigments by t.l.c. and for their structure elucidation is presented. Separated bile pigments are characterized by t.l.c. of derived dipyrrolic azopigments. 2. At the tetrapyrrolic stage hydrolysis in strongly alkaline medium followed by t.l.c. demonstrates the presence of bilirubin-IIIalpha, -IXalpha and -XIIIalpha and allows assessment of their relative amoun...
متن کاملSomatostatin inhibits the effect of secretin on bile flow and on hepatic bilirubin transport in the rat.
Increasing amounts of porcine secretion (0.05 to 2.00 clinical units/h/100 g body wt) given to rats during a continuous infusion of bilirubin, increased bile flow and the apparent maximal biliary excretion of bilirubin ('Tm'). This increment was caused by an enhanced biliary output of bilirubin monoconjugates. The effect was dose dependent but maximal at a secretin infusion of 0.80 CU. Somatost...
متن کاملChanges in bilirubins in human prenatal development.
1. A densitometric method has been developed for the quantification of azodipyrroles derived from dog bile pigments treated with diazotized ethyl anthranilate. 2. This method was used to estimate the bilirubins in bile and meconium from foetuses of 14-36 weeks gestation. 3. The proportion of the bilirubins in foetal bile changed during gestation. (a) No bile pigments were found until 14 weeks. ...
