چکیده

Heparan sulfate proteoglycans are ubiquitously found at the cell surface and extracellular matrix in all the animal species. This review will focus on the structural characteristics of the heparan sulfate proteoglycans related to protein interactions leading to cell signaling. The heparan sulfate chains due to their vast structural diversity are able to bind and interact with a wide variety of proteins, such as growth factors, chemokines, morphogens, extracellular matrix components, enzymes, among others. There is a specificity directing the interactions of heparan sulfates and target proteins, regarding both the fine structure of the polysaccharide chain as well precise protein motifs. Heparan sulfates play a role in cellular signaling either as receptor or co-receptor for different ligands, and the activation of downstream pathways is related to phosphorylation of different cytosolic proteins either directly or involving cytoskeleton interactions leading to gene regulation. The role of the heparan sulfate proteoglycans in cellular signaling and endocytic uptake pathways is also discussed.

Impact of SULF1 Gene on Angiogenesis

Single-gene disorders occur when mutation in a gene causing alteration of gene function while in multifactorial disorders, mutations occur in multiple genes, and these are usually coupled with environmental causes. In addition, in a multifactorial disorder such as diabetes, the complication is under the influence of different genes. For example, in diabetic retinopathy many genes are involved i...

Role of heparan sulfate proteoglycans in cell signaling and cancer.

The correct growth and development of multicellular organisms depends on the reception of numerous extracellular signals that activate various signal transduction cascades within the target cells. The activity of these pathways, such as the receptor tyrosine kinases (RTKs), transforming growth factor-β (TGF-β), Wnts, and Hedgehogs (Hh), are usually regulated by the binding of extracellular liga...

Cerebellar proteoglycans regulate sonic hedgehog responses during development.

Sonic hedgehog promotes proliferation of developing cerebellar granule cells. As sonic hedgehog is expressed in the cerebellum throughout life it is not clear why proliferation occurs only in the early postnatal period and only in the external granule cell layer. We asked whether heparan sulfate proteoglycans might regulate sonic hedgehog-induced proliferation and thereby contribute to the spec...

FGFs, heparan sulfate and FGFRs: complex interactions essential for development.

Fibroblast growth factors (FGFs) comprise a large family of developmental and physiological signaling molecules. All FGFs have a high affinity for the glycosaminoglycan heparin and for cell surface heparan sulfate proteoglycans. A large body of biochemical and cellular evidence points to a direct role for heparin/heparan sulfate in the formation of an active FGF/FGF receptor signaling complex. ...

Syndecans in tumor cell adhesion and signaling

Anchorage of cells to "heparin"--binding domains that are prevalent in extracellular matrix (ECM) components is thought to occur primarily through the syndecans, a four-member family of transmembrane heparan sulfate proteoglycans that communicate environmental cues from the ECM to the cytoskeleton and the signaling apparatus of the cell. Known activities of the syndecans trace to their highly c...
