WASP, Tregs, and food allergies - rare disease provides insight into a common problem.

چکیده
Dysregulation of the type 2 immune system presents with various manifestations, including allergic inflammation, and has emerged as an alarming public health issue. The pathological mechanisms that underlie T helper type 2 cell-driven (Th2-driven) allergic diseases remain unclear. In particular, it is not completely understood how type 2 immunity is restricted in inflammatory responses. In this issue of the JCI, Lexmond et al. use Wiskott-Aldrich syndrome as a model disease and establish that the Wiskott-Aldrich gene product (WASP) serves an essential role in T regulatory cells to contain Th2 effector cell differentiation and prevent allergic sensitization to dietary antigens.

Moral Considerations in Pediatric Food Allergies
Food allergies are common health problem among children. They carry a significant risk of severe allergic reactions. These disorders are chronic conditions in which the immune system becomes hypersensitive to some food products. It is estimated that 8% of children under the age of three have a type of food allergy. The common allergenic foods include cow’s milk, wheat, peanuts, egg, soy and fis...
متن کاملFOXP3+ Tregs require WASP to restrain Th2-mediated food allergy.
In addition to the infectious consequences of immunodeficiency, patients with Wiskott-Aldrich syndrome (WAS) often suffer from poorly understood exaggerated immune responses that result in autoimmunity and elevated levels of serum IgE. Here, we have shown that WAS patients and mice deficient in WAS protein (WASP) frequently develop IgE-mediated reactions to common food allergens. WASP-deficient...
متن کاملPrevalence of Allergies and Food Intolerance: a Comparison Between the Persian Community and Canadians of European Descent
Background: The prevalence of reported allergic diseases is higher in North-America than in other countries around the world. Further, certain types of allergies are more common in some geographic regions or amongst different ethnic populations. This phenomenon has not been well studied amongst the Persian population in the Canadian context; therefore, the current study aimed to compare the pre...
متن کاملWiskott-Aldrich syndrome protein is required for regulatory T cell homeostasis.
Wiskott-Aldrich syndrome protein (WASp) is essential for optimal T cell activation. Patients with WAS exhibit both immunodeficiency and a marked susceptibility to systemic autoimmunity. We investigated whether alterations in Treg function might explain these paradoxical observations. While WASp-deficient (WASp(-/-)) mice exhibited normal thymic Treg generation, the competitive fitness of periph...
متن کاملCeliac Disease
Celiac disease also known as gluten-sensitive enteropathy is characterized by intestinal mucosal damage and malabsorption from dietary intake of wheat, rye or barley. Symptoms may appear with introduction of cereal in the first 3 years of life. A second peak in symptoms occurs in adults during the third or forth decade and even as late as eight decade of life. The prevalence of this disease is ...
