چکیده
Three isotoxins (SP I-III) of the ,-bungarotoxin family were purified to homogeneity via a series of isolation procedures including a final step of h.p.l.c. on an SP column washed with a linear gradient of 0.2-0.6 M sodium acetate at pH 7.4. Their proportions varied greatly with the batch ofvenom. Each isotoxin was demonstrated by SDS/PAGE to contain a phospholipase A2 subunit and a non-phospholipase A2 subunit. The three proteins were reductively alkylated with 4-vinylpyridine and the alkylated derivatives of the two subunits of each isotoxin were separated. N-Terminal sequence analysis of the alkylated derivatives revealed that the three isotoxins probably share a common phospholipase A2 subunit but differ in their non-phospholipase A2 subunits. The non-phospholipase A2 subunits of SP II and SP III were identical with those of f32and Al-toxin respectively, except that there was an additional valine inserted between

Met-8 of the beta 1-bungarotoxin phospholipase A2 subunit is essential for the phospholipase A2-independent neurotoxic effect.
beta 1-Bungarotoxin consists of a phospholipase A2 subunit and a non-phospholipase A2 subunit. The toxin was oxidized with a 100-fold molar excess of chloramine T with respect to the methionine content of the protein in 0.1 M Tris/HCl at pH 8.5 and at room temperature. Reactivities of the two methionine (Met-6 and Met-8 of the phospholipase A2 subunit), five histidine, 14 tyrosine and one trypt...
متن کاملAnalysis of nociceptive effects of neurotoxic phospholipase A2 from Vipera nikolskii venom in mice
Phospholipases A2 are represented in snake venoms by several types and possess diverse biological activities including neurotoxicity. Previously, we isolated and characterized two neurotoxic phospholipases A2 (HDP-1 and HDP-2) from the venom of Nikolski's viper (Vipera nikolskii), which were heterodimers composed of two non-covalently bound subunits. Each heterodimer consisted of an enzymatical...
متن کاملThe Effect of Eight Weeks of High-intensity Interval Training on Lipoprotein-associated Phospholipase A2 and Lipid Profile in a Male Rat Model of Type 2 Diabetes
Introduction: Type 2 diabetes (T2D) causes hyperglycemia, hyperinsulinemia, and dyslipidemia, which are all risk factors for atherosclerosis. Lipoprotein-associated phospholipase A2 (Lp-PLA2) has been recognized as an indicator of atherosclerosis due to its role in vessel inflammation. This study aimed to investigate the effect of high-intensity interval training (HIIT) on serum levels of Lp-PL...

Abstract Background: The Beijing strain of Mycobacterium tuberculosis is responsible for more than 25% of cases of Tuberculosis in the world, it has a high transmission potential and there is a significant correlation between Beijing strain and multidrug resistance. In this study we compared the presence of Phospholipase C genes in Beijing and Nonbeijing strains of Mycobacterium tuberculosis...
متن کاملP 25: The Facilitatory Action of Snake Venom Phospholipase A2 Neurotoxins by Which Increase the Release of Acetylcholine, May Improve Alzheimer\'s Disease Symptoms
Introduction: In a serious brain disorder like Alzheimer's disease, the levels of acetylcholine (Ach) drop significantly. The gradual death of cholinergic brain cells leads to a profound loss of memory and learning ability. Acetylcholine is the chemical messenger that sends messages from one neuron to another in the area of the brain used for memory. Many of the current medications act to enhan...
