چکیده

Introduction Recombinant DNA techniques have revolutionized the field of microbiology. The techniques are quite helpful to analyse and modify various functions of cells. Using recombinant DNA techniques we have succeeded in analysing and modifying the metabolic functions of micro-organisms [ l]. Focusing on the glyoxalase system we have analysed a glycolytic-methylglyoxal cycle and have produced a ‘jumbo’ yeast, which could produce a large amount of S-lactoylglutathione [2-41.

Inhibitors of glyoxalase I: design, synthesis, inhibitory characteristics and biological evaluation.

Glyoxalase I (EC 4.4.1 .S lactoylglutathione lyase) catalyses the formation of S-u-lactoylglutathione from methylglyoxal and glutathione [ 11. The molecular species that binds to the active site of glyoxalase I is the hemithioacetal, formed nonenzymically from methylglyoxal and reduced glutat hione, y-L-glutamyl-S-( 1 -hydroxy-2-oxopropyl)L-cysteinylglycine. The isomerization of the hemithioace...

Bacterial glyoxalase I enzymes: structural and biochemical investigations.

A number of bacterial glyoxalase I enzymes are maximally activated by Ni2+ and Co2+ ions, but are inactive in the presence of Zn2+, yet these enzymes will also bind this metal ion. The structure-activity relationships between these two classes of glyoxalase I serve as important clues as to how the molecular structures of these proteins control metal-activation profiles.

Glyoxalase I--structure, function and a critical role in the enzymatic defence against glycation.

Glyoxalase I is part of the glyoxalase system present in the cytosol of cells. The glyoxalase system catalyses the conversion of reactive, acyclic alpha-oxoaldehydes into the corresponding alpha-hydroxyacids. Glyoxalase I catalyses the isomerization of the hemithioacetal, formed spontaneously from alpha-oxoaldehyde and GSH, to S -2-hydroxyacylglutathione derivatives [RCOCH(OH)-SG-->RCH(OH)CO-SG...

The effect of fasting on the important molecular mechanisms related to cancer treatment

Fasting does have remarkable benefits in the treatment of cancer and another diseases such as metabolic syndrome, diabetes, and a multitude of other chronic diseases. It has been determined that fasting could play an important role during cancer treatment and progression via the regulation of insulin-like growth factor-1 (IGF-1) as well as other growth factors. Also, it has been shown that fast...

A stress-responsive glyoxalase I from the parasitic nematode Onchocerca volvulus.

Glyoxal, methylglyoxal and other physiological alpha-oxoaldehydes are formed by the lipid peroxidation, glycation and degradation of glycolytic intermediates. They are detoxified enzymically by the glyoxalase system. To investigate the physiological function of glyoxalase I in parasitic organisms, the cDNA for glyoxalase I from the filarial nematode Onchocerca volvulus (designated Ov-GloI) has ...
