An Improved Method for the Measurement of Urinary 17-ketosteroids of Small Laboratory Animals.

چکیده
formula (1, 2) has circumvented this problem to some extent. However, the amount and type of interfering chromogen in urine is not a constant factor, so that the routine use of the correction formula often leads to unreliable results. Recently Sheath (3) reported that the addition of formalin to urine prior to hot-acid hydrolysis greatly reduced the formation of nonsteroidal chromogens. Also, the partitioning of the Zimmerman color and nonspecific chromogens between methylene chloride and aqueous ethanol prior to colorimetry has been described (4-6). The combination of these two technics in our laboratory has led to an assay of urinary 17-ketosteroids of small laboratory animals with a degree of reliability and accuracy not obtainable with either technic alone.

Test-tube extraction method for the micro-determination of urinary 17-ketosteroids.
SIMPLE, RAPID TEST-TUBE EXIRACTION TECHNIC for the determination of urinary 17-ketosteroids has seen developed in our laboratories. A complete urinary 17-ketosteroid determination was carried out in a glass stoppered photometer cuvet by combining this extraction technic with a colorimetric method. Three colorimetric methods, Zimmermann (1), (2), Pincus (3), and Cohen and Kaluszyner (4) were com...

hydrochloride (10 mg three times a day for as long as 20 days) did not affect values for 17-hydroxycorticosteroids. In vitro data do show that there is a small increase in the apparent value for urinary 17-hydroxycorticosteroids when chlordiazepoxide or its lactam or N-despiethyl metabolite is added to urine. However, this increase does not appear to be large enough to be of significance. The f...
متن کاملA rapid method for the estimation of urinary 17-ketosteroids.
HE QUANTITATIVE ESTIMATION of the urinary 17-ketosteroids in the routine laboratory still presents a number of problems. The most serious of these is the presence of nonketonic chronlogens in tile final extracts prepared by the usual extraction methods. Tile removal of these interfering substances by tile application of Girard’s reagent (1, 2) is not feasible for the routine clinical laboratory...
متن کاملTotal neutral 17-ketosteroids; clinical method for measurement.
ITH INCREASING DEMAND for the determination of urinary total 17-ketosteroids as a diagnostic tool, a need has developed for a , rapid method suitable for clinical use. Such a method was pubby Drekter et al. (1) and modified by Masuda and Thuline (2). We further modified the latter method to permit a larger number of s to be run at one time. We have substituted equipment which is akable and less...
متن کاملUrinary glucocorticoid excretion.
In recent years two comparatively simple methods have been described for the assessment of urinary glucocorticoid excretion. Reddy, Jenkins, and Thorn in 1952 gave details of a method for estimating urinary " 17-hydroxycorticoids." This involved extraction of the urinary glucocorticoids (cortisone and hydrocortisone) and their metabolites with n-butyl alcohol and the use of the Porter-Silber co...
