چکیده

Mice lacking the dopamine transporter (DAT-/-) are characterized by high extracellular dopamine levels and spontaneous hyperlocomotion. We performed a detailed analysis of the behavioral phenotype of DAT-/mice in order to identify other behavioral impairments associated with the hyperdopaminergic tone of these mutant mice. In particular, we investigated locomotor activity, exploration, social and maternal behaviors, which are known to be regulated by dopamine. DAT-/mice were easily aroused by novelty and always responded with an hyperlocomotion, which interfered with habituation to the testing environment, exploratory behavior in an open field and coping response to forced swimming stress. Social behaviors such as interaction with an unknown congener or aggressiveness were not modified in DAT-/mice compared to DAT+/and DAT+/+ mice, although the maternal behavior of the mutant females was severely disturbed. Haloperidol and clozapine reversed hyperactivity in DAT-/mice with a rightward shift of the dose-response curve compared to control animals, suggesting a dopamine-mediated effect. These results emphasize the role of dopamine regulation in locomotion, exploration, and maternal behaviors and suggest that mice with a genetic deletion of the DAT may represent an useful model t o elucidate the altered behavioral processes accompanying pathological conditions associated with hyperdopaminergic function.

Persistent Hyperdopaminergia Decreases the Peak Frequency of Hippocampal Theta Oscillations during Quiet Waking and REM Sleep

Long-term changes in dopaminergic signaling are thought to underlie the pathophysiology of a number of psychiatric disorders. Several conditions are associated with cognitive deficits such as disturbances in attention processes and learning and memory, suggesting that persistent changes in dopaminergic signaling may alter neural mechanisms underlying these processes. Dopamine transporter knocko...

Hyperdopaminergia and NMDA receptor hypofunction disrupt neural phase signaling.

Neural phase signaling has gained attention as a putative coding mechanism through which the brain binds the activity of neurons across distributed brain areas to generate thoughts, percepts, and behaviors. Neural phase signaling has been shown to play a role in various cognitive processes, and it has been suggested that altered phase signaling may play a role in mediating the cognitive deficit...

Dopamine transporter knockout mice in experimental neuropharmacology

OF TALK The monoaminergic neurotransmitter dopamine (DA) has been implicated in multiple brain disorders including schizophrenia, attention deficit hyperactivity disorder (ADHD), addiction and Parkinson's disease. Dopamine transporter (DAT) is a major regulator of both the intensity of extracellular dopamine (DA) signaling and presynaptic neuronal homeostasis. By deleting the gene encoding the ...

Recombinant Adeno-Associated Virus-mediated rescue of function in a mouse model of Dopamine Transporter Deficiency Syndrome

Dopamine Transporter Deficiency Syndrome (DTDS) is a rare autosomal recessive disorder caused by loss-of-function mutations in dopamine transporter (DAT) gene, leading to severe neurological disabilities in children and adults. DAT-Knockout (DAT-KO) mouse is currently the best animal model for this syndrome, displaying functional hyperdopaminergia and neurodegenerative phenotype leading to prem...
