The specificity of tryptophan analogues as inducers, substrates, inhibitors, and stabilizers of liver tryptophan pyrrolase.

چکیده
In animals, liver tryptophan pyrrolase was induced independently by administration of its substrate L-tryptophan (I), and of the hormone hydrocortisone (2). Some analogues of tryptophan which are not substrates of the tryptophan pyrrolase, such as c+mcthyl-DL-tryptophan (3) and D-tryptophan,’ have been reported to induce the enzyme in intact animals. Since a substance may cause enzyme induction in viva by stressinduced adrenal secretion and the hormone-induced mechanism, adrenalectomized rats were used to determine the specificity of the substrate-induced enzyme response. The same analogues of tryptophan also were tested as substrates, inhibitors, and stabilizers of the cell-free enzyme preparation to determine whether there was any relation between the inducing activity of a substance and its affinity for the enzyme.

Effect of cytoplasmic particles on tryptophan pyrrolase activity of rat liver.
An activation of tryptophan pyrrolase in vitro distinct from the previously known conversion of inactive apotryptophan pyrrolase to active holotryptophan pyrrolase by hematin or by boiled particulate fractions of liver is described. This stimulation occurs by the addition of small amounts of mitochondria or microsomes to cell sap in the presence of excess methemoglobin. The particulate activato...
متن کاملTryptophan pyrrolase of liver. II. The activating reactions in crude preparations from rat liver.
The activation of the apoenzyme of tryptophan pyrrolase in preparations from hydrocortisone-induced rats was shown to consist of two separate, sequential steps, only the second of which is necessary to activate preparations from tryptophan-induced rats. These reactions are as follows. (a) Conjugation of the apoenzyme to the oxidized holoenzyme with hematin supplied by added methemoglobin, a rea...
متن کاملThe Roles of Synthesis and Degradation in the Control of Rat Liver Tryptophan Pyrrolase.
It is now well established that in animal tissues a large number of enzyme activities can be increased by the administration of hormones (l-3), by specific substrates (1, 4), or by alterations in the nutritional state of the animal (58). In certain of these cases an increased content of newly synthesized enzyme protein has been shown to account for the increases in enzyme activity, as opposed t...
متن کاملTryptophan Catabolism by Tryptophan Pyrrolase in Rat Liver THE EFFECT OF TRYPTOPHAK LOADS AKD CHAKGES IN TRYPTOPHAN PYRROLASE ACTIVITY*
We investigated how changes in tryptophan pyrrolase activity and tryptophan loads affect the breakdown of tryptophan by tryptophan pyrrolase. Breakdown of tryptophan was estimated by injecting rats with [ring-ZJ%]tryptophan and measuring respiratory 14C02. We concluded, contrary to previous reports, that induction of tryptophan pyrrolase definitely will increase the rate of tryptophan breakdown...
متن کاملThe activation and induction of rat liver tryptophan pyrrolase in vivo by its substrate.
Parenteral administration of tryptophan has been shown to increase markedly rat liver tryptophan pyrrolase activity (1). This phenomenon is often referred to as induction or adaptation due to its similarity to substrate-induced enzyme formation in microorganisms. The preceding article (2) documents the presence of a factor in microsomes which activates tryptophan pyrrolase of normal liver cell ...
