چکیده

Introduction HIV transmission occurs predominantly across mucosal surfaces. An ideal preventive strategy would be to target HIV at mucosal entry sites to prevent infection. We developed a novel epithelial stem cells-based AIDS preventive approach in female macaques. This approach is based on the ability of therapeutic lentiviral vectors integrated in mucosal epithelial stem cells to induce virusspecific cellular immune responses at mucosal sites of viral entry. We first intended to expose the cervicovaginal tract to conditions used for intravaginal vaccine delivery and non-invasively image local tissues to determine the vaccination effects on epithelial integrity and generation of antigen-specific mucosal immune responses.

A neonatal Fc receptor-targeted mucosal vaccine strategy effectively induces HIV-1 antigen-specific immunity to genital infection.

Strategies to prevent the sexual transmission of HIV include vaccines that elicit durable, protective mucosal immune responses. A key to effective mucosal immunity is the capacity for antigens administered locally to cross epithelial barriers. Given the role of neonatal Fc receptor (FcRn) in transferring IgG across polarized epithelial cells which line mucosal surfaces, FcRn might be useful for...

Gene Delivery to Mesenchymal Stem Cells

There is increasing trend in using recombinant stem cells as novel therapeutic candidates in different diseases. These studies encompass different applications from targeted homing of Mesenchymal Stromal (stem) Cells (MSC), to arming them with different cytokines. Resistance to transfection or transduction methods had urged researchers to look for better gene delivery alternates and optimizing ...

Expression of Cytokeratin 19 in the epithelial cell of Azo-exposed buccal mucosa

Background: Azo is a synthetic dye used in batik industries. It can be toxic to the tissue when exposed via inhalation, swallowing, or direct contact. Expression of cytokeratin will change in hyperplastic and cancer of the oral mucosa. Expression of Cytokeratin 8, 18, 19 is strong in the epithelial cells that undergo excessive hyperproliferation and oral mucosal changes in le...

Pluripotent stem cells as the cells with a capacity for self-renewal and differentiation into various specificcell types have been highly regarded in regenerative medicine studies. To repair the eye disease damages, thedifferentiation into retinal pigment epithelial cells of pluripotent stem cells has gained great importance inrecent decades because the inappropriate function of these cells is ...

Mayer, Lloyd. Current Concepts in Mucosal Immunity. I. Antigen presentation in the intestine: new rules and regulations. Am. J. Physiol. 274 (Gastrointest. Liver Physiol. 37): G7–G9, 1998.—The recognition that immune responses in the intestine differ from those seen systemically has led to a search for novel pathways involved in mucosal immunoregulation. One cell type that has surfaced as a pri...
