چکیده

Ethics Statement Production of specific pathogen–free eggs (VALO BioMedia GmbH, OsterholzScharmbeck, Germany) was performed in accordance with the guidelines of the European Pharmacopoeia (EP7.0.5.2.2.) and the US Department of Agriculture Veterinary Services (Memorandum 800.65). All procedures regarding embryonated chicken eggs were in accordance with the German Animal Protection Law. For infection, fertilized chicken eggs at embryonation day 11 were inoculated with virus into the allantois sack or onto the chorioallantoic membrane. Development of embryos was terminated at the 17th day of embryonation by cooling the eggs overnight at 4°C. No further specific approval is needed for experiments on embryonated birds before the time of hatching.

Development of Multiplex Reverse Transcription-Polymerase Chain Reaction for Simultaneous Detection of Influenza A, B and Adenoviruses

Background and objective:Millions of people in developing countries lose their lives due to acute respiratory infections, such as Influenza A & B and Adeno viruses. Given the importance of rapid identification of the virus, in this study the researchers attempted to design a method that enables detection of influenza A, B, and adenoviruses, quickly and simultaneously. ...

Protocol for Metagenomic Virus Detection in Clinical Specimens1

Sixty percent of emerging viruses have a zoonotic origin, making transmission from animals a major threat to public health. Prompt identification and analysis of these pathogens are indispensable to taking action toward prevention and protection of the affected population. We quantifiably compared classical and modern approaches of virus purification and enrichment in theory and experiments. Ev...

Optimization and validation of sample preparation for metagenomic sequencing of viruses in clinical samples

BACKGROUND Sequence-specific PCR is the most common approach for virus identification in diagnostic laboratories. However, as specific PCR only detects pre-defined targets, novel virus strains or viruses not included in routine test panels will be missed. Recently, advances in high-throughput sequencing allow for virus-sequence-independent identification of entire virus populations in clinical ...

Use of RNALater® Preservation for Virome Sequencing in Outbreak Settings

Outbreaks of infectious diseases may occur in animal and human populations; this underlines the need for suitable One Health approaches. During outbreak situations, straightforward identification of etiological agents is indispensable for taking countermeasures. A recently published protocol for metagenomic virus detection in clinical specimens (TUViD-VM) was developed for snap-frozen tissues w...

Detection of avian influenza virus of H9 subtype in the feces of experimentally infected chickens by RT–PCR

Avian Influenza (AI) is a viral respiratory disease of domestic and wild birds. In the diagnostic laboratory, it is essential to have methods for rapid detection of avian respiratory viruses. Cloacal swabs collected from chickens experimentally infected with H9 subtype AI virus, used in a reverse transcription-polymerase chain reaction (RT-PCR) assay for detection of AI. In infected animals, ...
