From neurotrophins to immunotrophins

چکیده
The series of international conferences on nerve growth factor (NGF) and related molecules arose from a meeting held in 1986, to mark Rita Levi-Montalcini’s 77th birthday, and in recognition of the fact that her discovery of NGF opened up the field of trophic factors. One of the best characterized families of such trophic factors is the NGF family of neurotrophins, which in mammals also includes BDNF (brain derived neurotrophic factor), NT3 (neurotrophin 3) and NT4 (neurotrophin 4). The meeting has since become a biennial event held in alternating years with the Gordon Conference on neurotrophic factors. This year’s meeting, the seventh in the series, was held in the lovely Italian town of Modena and was honored once again by the presence of the Nobel laureate. In accordance with Levi-Montalcini’s views on expanding roles for neurotrophins outside the nervous system, the organizers prepared a wide-ranging program encompassing neuronal, non-neuronal and clinical aspects of NGF research. Presentations included some quite striking research topics, such as the effects of increased gravity on tissue levels of neurotrophins (D. Santucci, Rome, Italy), as well as more conventional studies on different aspects of neurotrophic factor biology. In this short report we focus on molecular and mechanistic advances presented at the meeting. We apologize to the many speakers and poster presenters whose data could not be highlighted due to space constraints. For a more thorough discussion of the neurotrophins and their actions, readers are referred to more comprehensive reviews (Bibel and Barde, 2000; Huang and Reichardt, 2001).

Activation of neurotrophins in lumbar dorsal root probably contributes to neuropathic pain after spinal nerve ligation
Objective(s): Neurotrophins (NTs) exert various effects on neuronal system. Growing evidence indicates that NTs are involved in the pathophysiology of neuropathic pain. However, the exact role of these proteins in modulating nociceptive signaling requires being defined. Thus, the aim of this study was to evaluate the effects of spinal nerve ligation (SNL) on NTs activation in the lumbar dorsal ...

Objective(s): Studies have confirmed that microgravity, as a mechanical factor, influences both differentiation and function of mesenchymal stem cells. Here we investigated the effects of simulated microgravity on neural differentiation of human adipose-derived stem cells (ADSCs). Materials and Methods:We have used a fast rotating clinostat (clinorotation) to simulate microgravity condition. R...
متن کاملThe Response of Skeletal Muscle-Expressed Neurotrophins to Acute Resistance Exercise in Male Wistar Rats
Background. BDNF and NT-4/5 have been proposed to be involved in the coordinated adaptations of the neuromuscular system to the elevated level of activity, but an activity-dependent expression of neurotrophins in skeletal muscle is not well established. Objectives. We, therefore, investigated the effect of one session of resistance exercise on mRNA expression of some neurotrophins in Slow and ...
متن کاملNeurotrophins in the developing and regenerating visual system.
The neurotrophins NGF, BDNF, NT-3 and NT-4 have a wide range of effects in the development and regeneration of neural circuits in the visual system of vertebrates. This review focuses on the localization and functions of neurotrophins in the retina, lateral geniculate nucleus, suprachiasmatic nucleus, superior colliculus/optic tectum, and isthmic nuclei. Research of the past 20 years has shown ...
متن کاملUsing transient transfected Chinese hamster ovary (CHO) cells by pET28a-LIC-NTF4 in gene therapy
Introduction: Neurotrophins, as a family of proteins, are responsible for induction of the survival, development, and function of neurons. Also, neurotropic factors are growth factors like Neurotrophins that help neurons survive. Moreover, Neurotrophins differentiate between progenitor cells so that neurons are formed. Despite the fact that the majority of mammalian brain neurons are produced p...
