Molecular breeding of Clostridia for butanol production from rice straw

چکیده

Biobutanol produced by Clostridium spp. can be used as a biofuel and/or chemical feedstock. For cost-effective and sustainable production of biobutanol, an untapped biomass such as rice straw could be used as a substrate. Rice straw, which is composed of cellulose, hemicellulose, and lignin, is cheap and abundant. However, the utilization of rice straw is limited because butanol-producing clostridia cannot degrade cellulose, which is the major carbon source in lignocellulose. To achieve butanol production from lignocellulose, cellulose needs to be hydrolyzed to dior monosaccharides that can then be metabolized by Clostridium spp. In this study, we developed a system for butanol production from rice straw using a co-culture of cellulose-degrading Clostridium thermocellum and butanol-producing Clostridium saccharoperbutylacetonicum. In addition, we showed that β-glucosidase enhances butanol production from rice straw, and identified promoters that may be used for high-level expression of the β-glucosidase gene to increase productivity.

Utilization of Urea and Urease Treated Rice Straw by Indigenous Growing Cattle

The experiment was conducted to study the effect of treatment of rice straw with urea and a urease containing midden soil on the chemical composition of treated rice straw, feed intake of the animals, nutrients digestibility, body weight gain, feed conversion efficiency and overall economy of feeding for a period of 105 days. Twelve indigenous growing cattle (live weight 130.00±1.67 kg) were se...

In the current study, a set of 96 recombinant inbred lines (RIL) at F8, derived from a cross between two varieties, Anbarbu (sensitive to drought stress) and Sepidroud (tolerant to drought stress) were used. The experiment was performed at Gonbad Kavous located in the Golestan province of Iran in 2010–2011 using two augmented designs at normal and stress conditions, separately. The RIL populati...

Development of semi-defined rice straw-based medium for butanol production and its kinetic study

Rice straw is one of the potential economic feedstock for biobutanol production through ABE fermentation. However, the rice straw hydrolysate-based fermentation medium needs to be supported with nutritional elements. In this study, an attempt is made to optimize the rice straw hydrolysate-based fermentation medium employing Clostridium acetobutylicum MTCC 481 using Taguchi design of experiments...

Bacterial populations colonizing and degrading rice straw in anoxic paddy soil.

Rice straw is a major substrate for the production of methane, a greenhouse gas, in flooded rice fields. The bacterial community degrading rice straw under anoxic conditions was investigated with molecular methods. Rice straw was incubated in paddy soil anaerobically for 71 days. Denaturing gradient gel electrophoresis (DGGE) of the amplified bacterial 16S rRNA genes showed that the composition...

Evaluation of biobutanol production from non-pretreated rice straw hydrolysate under non-sterile environmental conditions.

The study aims to investigate a cost-effective approach to convert non-pretreated rice straw hydrolysate into biobutanol. The influences of the initial cell concentration and incubation temperature on biobutanol production were evaluated under both sterile and non-sterile conditions. Results indicate that 100% glucose utilization could be achieved for initial cell concentrations greater than 21...
