The role of poly (ADP-ribose) polymerase in ventilator-induced lung injury

چکیده
Some issues have not been addressed appropriately. The authors show increased levels of TNFα in lung homogenate after 2 hours of lung-protective ventilation (LPV). Previous data from our laboratory have shown in the healthy mouse lung that so-called protective mechanical ventilation (tidal volume, 8 ml/kg; peak airway pressure, 10 to 12 cmH2O; positive end-expiratory pressure, 4 cmH2O) induces a pulmonary inflammatory response [2]. In addition to elevated levels of TNFα, we found increased expression of IL-1β, IL-6, and keratinocyte-derived chemokine in the lung homogenate and found an increased number of pulmonary leucocytes in mice mechanically ventilated for 2 hours. Electron microscopy revealed evidence for type I pneumocyte membrane disruption and endothelial detachment, indicating structural injury. In line with the findings by Kim and colleagues, the wet/dry ratio was not affected after 2 hours of mechanical ventilation – although in our study 4 hours of protective ventilation did increase the wet/dry ratio.

Pulmonary and renal protection: targeting PARP to ventilator-induced lung and kidney injury?
Both acute lung injury and acute kidney injury (AKI) are frequent and serious problems in intensive care medicine. Therefore, the avoiding of any iatrogenic insult to these organs is of great importance. While an increasing body of evidence suggests that mechanical ventilation is capable of inducing lung and distant organ injury, the complex underlying molecular mechanisms remain insufficiently...
متن کاملInflammatory and transcriptional roles of poly (ADP-ribose) polymerase in ventilator-induced lung injury
INTRODUCTION Poly (ADP-ribose) polymerase (PARP) participates in inflammation by cellular necrosis and the nuclear factor-kappa-B (NF-kappaB)-dependent transcription. The purpose of this study was to examine the roles of PARP in ventilator-induced lung injury (VILI) in normal mice lung. METHODS Male C57BL/6 mice were divided into four groups: sham tracheostomized (sham), lung-protective venti...
متن کاملEffects of Poly (ADP-ribose) Polymerase Inhibition on DNA Integrity and Gene Expression in Ovarian Follicular Cells in Mice with Endotoxemia
Background: A mouse model of lipopolysaccharide (LPS)-induced inflammation was used to investigate the effect of pharmacological inhibition of nuclear enzyme PARP-1 on oocyte maturation, apoptotic and necrotic death, as well as DNA integrity of follicular cells. Also, the relative expression of cumulus genes (HAS2, COX2, and GREM1) associated with oocyte developmental competence was assessed. M...
متن کاملInhibition of poly(ADP-ribose) polymerase prevents allergen-induced asthma-like reaction in sensitized Guinea pigs.
Poly(ADP-ribose) polymerase (PARP) plays an important role in tissue injury in conditions associated with oxidative stress and inflammation. Because asthma is a chronic inflammatory disorder of the airways, we designed the present experimental study to evaluate the effects of PARP inhibition on allergen-induced asthma-like reaction in ovalbumin-sensitized guinea pigs. Cough and dyspnea in respo...
متن کاملAdrenomedullin in inflammatory process associated with experimental pulmonary fibrosis
BACKGROUND Adrenomedullin (AM), a 52-amino acid ringed-structure peptide with C-terminal amidation, was originally isolated from human pheochromocytoma. AM are widely distributed in various tissues and acts as a local vasoactive hormone in various conditions. METHODS In the present study, we investigated the efficacy of AM on the animal model of bleomycin (BLM)-induced lung injury. Mice were ...
