Purification and properties of phenylethanolamine-N-methyl transferase.

چکیده
The methylation of norepinephrine to epinephrine was first demonstrated by Biilbring with the use of minced adrenal gland and adenosine triphosphate (2). Soon after, Keller, Boissonnas, and du Vigneaud (3) showed that the methyl group of methionine is incorporated into the epinephrine of the adrenal gland in the intact animal. Subsequently, S-adenosylmethionine was shown to be the methyl donor in the N-methylation of norepinephrine to epinephrine (4). This report will describe the assay, purification, properties, and distribution of an enzyme, phenylethanolamine-N-methyl transferase, that N-methylates the nitrogen of norepinephrine as well as that of a number of phenylethanolamine derivatives.

Purification and properties of hydroxyindole-O-methyl transferase.
The demonstration of the hormone mclatonin (N-acetyl-Bmethoxytryptamine) in bovine pineal gland (1) prompted a study of the enzymes involved in its formation and metabolism. In preliminary studies (2, 3) we described an enzyme, hydroxyindole-O-methyl transferase, in the pineal gland of cows, that catalyzes the 0-methylation of N-acetyl-serotonin to melatonin. This paper describes the purificati...
متن کاملEndothelial cells are able to synthesize and release catecholamines both in vitro and in vivo.
Recently it has been demonstrated that catecholamines are produced and used by macrophages and mediate immune response. The aim of this study is to verify whether endothelial cells (ECs), which are of myeloid origin, can produce catecholamines. We demonstrated that genes coding for tyrosine hydroxylase, Dopa decarboxylase, dopamine β hydroxylase (DβH), and phenylethanolamine-N-methyl transferas...

متن کاملRegulation of catecholamine-synthesising enzymes and beta-adrenoceptors gene expression in ventricles of stressed rats.
Stress exposure activates the sympathoneural system, resulting in catecholamine release. Chronic stress is associated with development of numerous disorders, including cardiovascular diseases. Here we investigated the expression of mRNAs for catecholamine biosynthetic enzymes tyrosine-hydroxylase, dopamine-beta-hydroxylase and phenylethanolamine N-methyl-transferase, and for beta(1)- and beta(2...
