چکیده
Objective: The developmental process of vertebrate inner ear is complex. MicroRNAs (miRNAs) have been reported to be involved in the development of inner ear. The current study was designed to explore the potential role of miR-18a in the development of inner ear. Methods: The expression of miR-18a and lysine demethylase 5B (Kd5mb) in inner ear tissues and house Ear Institute-Organ of Corti 1 (HEIOC1) cells was detected by miRNA in situ hybridization (MISH) and immunofluorescence (IF), respectively. TargetScane was used to predict the potential target genes of miR-18a. Luciferase reporter assay, real-time PCR, and Western blot were performed to confirm that Kdm5b is a target gene of miR-18a. In addition, the effects of miR-18a aberrant expression on H3K4me3 level were detected by Western blot. Results: The results showed that the expression of miR-18a was gradually increased and the expression of Kdm5b was gradually decreased during the inner ear development. Kdm5b was confirmed as a target gene of miR-18a and was negatively regulated by miR-18a. Moreover, the results showed that the expression of H3K4me3 was enhanced by miR-18a mimic but weakened by miR-18a inhibitor. Conclusion: This study provides evidence that miR-18a and Kdm5b was dynamically regulated in the developmental process of inner ear. The effects of miR-18a on the inner cell development might be by directly targeting Kdm5b and indirectly regulating H3K4me3.

MicroRNAs are essential for development and function of inner ear hair cells in vertebrates.
MicroRNAs (miRNAs) inhibit the translation of target mRNAs and affect, directly or indirectly, the expression of a large portion of the protein-coding genes. This study focuses on miRNAs that are expressed in the mouse cochlea and vestibule, the 2 inner ear compartments. A conditional knock-out mouse for Dicer1 demonstrated that miRNAs are crucial for postnatal survival of functional hair cells...
متن کاملMicroRNA-194 Regulates the Development and Differentiation of Sensory Patches and Statoacoustic Ganglion of Inner Ear by Fgf4
BACKGROUND MicroRNA 194 is involved in the differentiation of various types of cells, such as adipose derived stem cells, human embryonic stem cells, and bone marrow mesenchymal stem cells. Previously, we found that miR-194 was highly expressed in the inner ear sensory patch and neurons in mice embryos. However, the role of miR-194 in the development of the inner ear and its underlying mechanis...

MicroRNA-200 family members are expressed in the developing mouse inner ear and in zebrafish (Danio rerio) olfactory epithelia, taste buds, and neuromasts, and have also been shown to be associated with differentiation of olfactory and taste buds. However, the role of the miR-200 family in the inner ear of zebrafish had not been studied. We investigated the expression and function of the miR-20...
متن کاملEffect of replacing cochlea contour with inner ear contour on cochlea dose-volume calculations in conventional 2 dimensional and conformal 3 dimensional radiotherapy of brain
Introduction: Sensorineural hearing loss (SNHL) is one of the possible complications of radiotherapy treatment of brain tumors. The auditory system of patients with brain tumors often is placed inside of radiation field and receives a significant amount of radiation dose resulting in hearing loss. The purpose of this study was to compare contouring and delivery dose to cochlea...
