HIV-1 Capsid as a Target for Antiviral Therapy

چکیده
Human immunodeficiency virus type 1 (HIV-1) is the causative virus responsible for acquired immune deficiency syndrome (AIDS), for which there is still no protective vaccine. Since 1983, more than 34 million people have been infected with HIV across the world, with 1.5 million people dying as a result of complications from infection each year. HIV-1 virions mainly infect cells of the immune system and lead to a gradual reduction in the proportion of CD4+ T cells [1]. This in turn causes a significant decrease in the patient’s ability to mount an immune response, and they die as a result of opportunistic infection. HIV-1 thus remains a major pathogenic threat to human health worldwide.

Combined Antiviral Therapy Using Designed Molecular Scaffolds Targeting Two Distinct Viral Functions, HIV-1 Genome Integration and Capsid Assembly
Designed molecular scaffolds have been proposed as alternative therapeutic agents against HIV-1. The ankyrin repeat protein (Ank(GAG)1D4) and the zinc finger protein (2LTRZFP) have recently been characterized as intracellular antivirals, but these molecules, used individually, do not completely block HIV-1 replication and propagation. The capsid-binder Ank(GAG)1D4, which inhibits HIV-1 assembly...
متن کاملHIV Capsid is a Tractable Target for Small Molecule Therapeutic Intervention
Despite a high current standard of care in antiretroviral therapy for HIV, multidrug-resistant strains continue to emerge, underscoring the need for additional novel mechanism inhibitors that will offer expanded therapeutic options in the clinic. We report a new class of small molecule antiretroviral compounds that directly target HIV-1 capsid (CA) via a novel mechanism of action. The compounds...
متن کاملTripeptide interference with human immunodeficiency virus type 1 morphogenesis.
Capsid assembly during virus replication is a potential target for antiviral therapy. The Gag polyprotein is the main structural component of retroviral particles, and in human immunodeficiency virus type 1 (HIV-1), it contains the sequences for the matrix, capsid, nucleocapsid, and several small polypeptides. Here, we report that at a concentration of 100 micro M, 7 of 83 tripeptide amides fro...
متن کاملIdentification of Vimentin as a Potential Therapeutic Target against HIV Infection
A combination of antiviral drugs known as antiretroviral therapy (ART) has shown effectiveness against the human immunodeficiency virus (HIV). ART has markedly decreased mortality and morbidity among HIV-infected patients, having even reduced HIV transmission. However, an important current disadvantage, resistance development, remains to be solved. Hope is focused on developing drugs against ce...
متن کاملRationally Designed Interfacial Peptides Are Efficient In Vitro Inhibitors of HIV-1 Capsid Assembly with Antiviral Activity
Virus capsid assembly constitutes an attractive target for the development of antiviral therapies; a few experimental inhibitors of this process for HIV-1 and other viruses have been identified by screening compounds or by selection from chemical libraries. As a different, novel approach we have undertaken the rational design of peptides that could act as competitive assembly inhibitors by mimi...
متن کاملSmall-molecule inhibition of human immunodeficiency virus type 1 infection by virus capsid destabilization.
Human immunodeficiency virus type 1 (HIV-1) infection is dependent on the proper disassembly of the viral capsid, or "uncoating," in target cells. The HIV-1 capsid consists of a conical multimeric complex of the viral capsid protein (CA) arranged in a hexagonal lattice. Mutations in CA that destabilize the viral capsid result in impaired infection owing to defects in reverse transcription in ta...
