Breakthrough Technologies Efficient Screening of Arabidopsis T-DNA Insertion Lines Using Degenerate Primers
نویسندگان
چکیده
The sequencing of the Arabidopsis plant genome is providing a fuller understanding of the number and types of plant genes. However, in most cases we do not know which genes are responsible for specific metabolic and signal transduction pathways. Analysis of gene function is also often confounded by the presence of multiple isoforms of the gene of interest. Recent advances in PCR-based reverse genetic techniques have allowed the search for plants carrying T-DNA insertions in any gene of interest. Here we report preliminary screening results from an ordered population of nearly 60,470 independently derived T-DNA lines. Degenerate PCR primers were used on large DNA pools (n 5 2,025 T-DNA lines) to screen for more than one gene family member at a time. Methods are presented that facilitated the identification and isolation of isoform-specific mutants in almost all members of the Arabidopsis H-proton ATPase gene family. Multiple mutant alleles were found for several isoforms.

متن کاملScreening insertion libraries for mutations in many genes simultaneously using DNA microarrays.
We describe a method to screen pools of DNA from multiple transposon lines for insertions in many genes simultaneously. We use thermal asymmetric interlaced-PCR, a hemispecific PCR amplification protocol that combines nested, insertion-specific primers with degenerate primers, to amplify DNA flanking the transposons. In reconstruction experiments with previously characterized Arabidopsis lines ...
متن کاملFusion primer and nested integrated PCR (FPNI-PCR): a new high-efficiency strategy for rapid chromosome walking or flanking sequence cloning
BACKGROUND The advent of genomics-based technologies has revolutionized many fields of biological enquiry. However, chromosome walking or flanking sequence cloning is still a necessary and important procedure to determining gene structure. Such methods are used to identify T-DNA insertion sites and so are especially relevant for organisms where large T-DNA insertion libraries have been created,...
متن کاملYeast Two Hybrid cDNA Screening of Arabidopsis thaliana for SETH4 Protein Interaction
SETH4 coding sequence with 2013 bp is a member of gene family expressed in gametophytic tissues of Arabidopsis thaliana. This fragment was PCR amplified using Kod Hi Fi DNA polymerase enzyme. This fragment was cloned into pGBKT7 bate vector and transformed E. coli DH5? cells containing vector were selected on LB medium containing Kanamycin. Finally, pGBKT7-SETH4 bate was transformed into yeast ...
متن کاملIsolation and molecular characterization of the RecQsim gene in Arabidopsis, rice (Oryza sativa) and rape (Brassica napus)
In any organism that reproduces sexually, DNA Recombination plays vital roles in the generation of allelic diversity as well as in preservation of genome fidelity. Genome fidelity is particularly important in plants because mutations occurring during the development of flowering plants are heritable and can be passed onto the next generation. One of the gene families that play crucial roles in ...
