Mechanism for Cotolerance in Nonlethally Conditioned Mixed Chimeras: Negative Selection of the V, T-cell Receptor Repertoire by Both Host and Donor Bone Marrow-Derived Cells

چکیده
Bone marrow (BM) chimeras prepared by complete recipient ablation (A-+B) exhibit donor-specific tolerance, yet survival is often limited by graft-versus-host disease (GVHD). Negative selection of potentially donor-reactive T cells, as assessed by relative T-cell receptor (TCRI-V, expression, is dependent on donor BM-derived eleting ligands. Mixed chimerism and tolerance for both donor and host antigens can be achieved using partial recipient myeloablation with 500 cGy total-body irradiation (TB11 before transplantation followed by cyclophosphamide (CyP) on day +2. To examine the influence of residual host elements on negative selection, the peripheral TCR-V, repertoire was analyzed in partially ablated C57BLIlOSnJ (B101 recipients reconstituted with BM from major histocompatibility complex (MHCI-disparate BlO.BR/SgSnJ or MHC, Hh-l and Mls-disparate

متن کاملHematopoietic cells and radioresistant host elements influence natural killer cell differentiation
Radioresistant host elements mediate positive selection of developing thymocytes, whereas bone marrow-derived cells induce clonal deletion of T cells with receptors that are strongly autoreactive. In contrast to T cell development, little is known about the elements governing the natural killer (NK) cell repertoire, which, similar to the T cell repertoire, differs between individuals bearing di...
متن کاملCross-species transplantation tolerance: rat bone marrow-derived cells can contribute to the ligand for negative selection of mouse T cell receptor V beta in chimeras tolerant to xenogeneic antigens (mouse + rat----mouse)
Mixed xenogeneic bone marrow reconstitution (mouse + rat----mouse) results in stable mixed lymphopoietic chimerism (1-48% rat), long-term survival, and the induction of stable functional donor-specific transplantation tolerance to xenoantigens in vivo. To examine the role of negative selection of potentially xenoreactive T lymphocytes during tolerance induction across a species barrier, mixed x...

متن کاملDefect in negative selection in lpr donor-derived T cells differentiating in non-lpr host thymus
Transplantation of bone marrow cells of lpr/lpr mice into irradiated normal mice fails to develop massive lymphadenopathy or autoimmunity but causes severe graft-vs.-host-like syndrome. To elucidate an abnormality of lpr/lpr bone marrow-derived T cells, we transplanted bone marrow cells of Mlsb lpr/lpr mice into H-2-compatible Mlsa non-lpr mice. Although lpr/lpr T cell precursors repopulated th...
