چکیده

BACKGROUND Paraoxonase 1 (PON1) is an ester hydrolase present in serum and in the liver. The aims of the present study were to investigate the following: (a) the relationship between serum PON1 activity alterations and the degree of liver damage in patients with chronic liver disease; (b) the influence of genetic variability on serum PON1 activity; and (c) the efficacy of serum PON1 activity measurement, alone and in combination with standard liver function tests, in the assessment of liver damage. METHODS We studied 68 patients with liver cirrhosis, 107 patients with chronic hepatitis, and 368 apparently healthy volunteers. Baseline and salt-stimulated PON1 activities were measured by the hydrolysis of paraoxon. PON1 genotyping at positions 55 and 192 was analyzed by PCR and restriction isotyping. RESULTS Baseline and stimulated PON1 activities were decreased (P <0.001) in chronic hepatitis and in liver cirrhosis. PON1 activity was significantly correlated with serum total proteins, albumin, and bilirubin in patients but not in controls. There were no significant differences with respect to allele and genotype frequencies between patients and controls. The combination of baseline serum PON1 with five standard biochemical tests had a higher classification accuracy (94% of patients; 96% of controls) than the five standard tests alone (75% of patients; 96% of controls). ROC plots demonstrated a high diagnostic accuracy for baseline serum PON1 [area under the curve, 0.89 (95% confidence interval, 0.86-0.93) in chronic hepatitis and 0.96 (95% confidence interval, 0.94-0.99) in cirrhosis]. Baseline PON1 provided the highest ROC area for cirrhosis vs controls. CONCLUSIONS The significant decrease of PON1 activity in chronic liver diseases is related to the degree of hepatic dysfunction and not to allelic or genotypic differences. Addition of serum PON1 activity measurement to the current battery of tests may improve the evaluation of chronic liver diseases.

Background: Non-alcoholic fatty liver disease (NAFLD) is a chronic liver disease is increasing in adults and children worldwide. Obesity, insulin resistance or diabetes type II, hyperlipidemia and hypertriglyceridemia plays a major role in the epidemiology of this disease. Evaluation of structure of paraoxonase (PON) and its relationship with various diseases

The paraoxonase family in humans includes the PON1, PON2, and PON3 genes. These genes are located on the long arm of chromosome 7 and are structurally similar .Between the nucleotide sequences of these three genes is about 70% and between their amino acid    sequences is about 60% compatibility. The three have 9 exons, which in the case of PON1 exists an addition code in position 106 (lysine) i...

IN VITRO EVALUATION OF ANTIOXIDANT CAPACITY OF THYMUS KOTSCHYANUS HYDRO-ALCOHOLIC EXTRACTS AND ITS EFFECT ON SERUM PARAOXONASE 1 ACTIVITY IN DIABETIC AND HEALTHY PERSONS

Background: Thymus species have significant amounts of phenolic and flavonoid compounds and demonstrate strong antioxidant activities. Paraoxonase1 act as antioxidant enzyme and protect the low-density lipoprotein against oxidation. In our study we aimed to evaluate the antioxidant capacity of Thymus Kotschyanus Hydroalcoholic extract and its effect on serum paraoxonase 1 activity in healthy an...

PARAOXONASE AND ARYLESTERASE ACTIVITY AMONG HYPERCHOLESTEROLEMIC PATIENTS

Human serum paraoxonase (PON) associated with high density lipoprotein (HDL) , has been postulated to have a role in protecting low-density lipoprotein (LDL) against oxidative damage, which has led to the proposal that PON is an anti-atherogenic and anti-inflammatory enzyme. It has genetic polymorphism at the 191 (R ->Q) and two alloenzymes A and B and three phenotypes A, B, and AB. We exa...

A new marker for lipid peroxidation: serum paraoxonase activity in non-alcoholic steatohepatitis.

BACKGROUND/AIMS Relationship between hepatic antioxidant paraoxonase 1 (PON1) activity, lipid peroxidation and liver injury was investigated in patients with non-alcoholic steatohepatitis. METHODS A total of 23 patients with non-alcoholic steatohepatitis (15 males, 8 females; mean age: 40.30+/-7.67 yrs) and 23 healthy controls (14 males, 9 females; mean age: 39.70+/- 8.78 yrs) were enrolled i...
