چکیده

Hearts isolated from 11and 12-day rat embryos were incubated in a simple salt solution to which was added the metabolic inhibitors iodoacetate, malonate, 2,4-dinitrophenol or trypan blue. Comparisons were made between the two age-groups and it was observed that both 11and 12-day hearts exhibited depressed contractile activity with the glycolytic inhibitor iodoacetate. Malonate did not depress contraction rate in the younger hearts but significantly depressed the rate in the older hearts. A greater inhibitory action on the older hearts as compared with the younger hearts was also produced by 2,4-dinitrophenol. These results were interpreted as further evidence of a shift in dependence on pathways other than glycolysis between days 11 and 12 of development. The teratogenic agent trypan blue was shown to depress contraction rate when added to the incubation medium. This effect could be reversed by adding glycolytic intermediates such as fructose-l,6-diphosphate or alpha glycerophosphate. These results were suggestive of a direct inhibitory effect of trypan blue either on glucose uptake or its complete catabolism.

Effects of hypothermia on energy metabolism in rat and Richardson's ground squirrel hearts.

Glycolysis, glucose oxidation, palmitate oxidation, and cardiac function were measured in isolated working hearts from ground squirrels and rats subjected to a hypothermia-rewarming protocol. Hearts were perfused initially for 30 min at 37 degrees C, followed by 2 h of hypothermic perfusion at 15 degrees C, after which hearts were rewarmed to 37 degrees C and further perfused for 30 min. Functi...

The effect of asafoetida essential oil on myocardial ischemic-reperfusion injury in isolated rat hearts

Objective: Previous studies reported that asafetida from Ferula assa-foetida Linn. species and its essential oil (AEO) have antioxidant effects. In the present study, the effect of AEO was evaluated on ischemic-reperfusion injury in isolated rat hearts. Materials and Methods: Forty-eight male Wistar rats were divided into 6 groups: 1) control group, 2) vehicle group, 3-5) AEO groups and, 6) car...

302 Metabolic Consequences and Predictabilit of Ventricular Fibrillation in Hypoxia

Background. Ventricular fibrillation has deleterious metabolic and functional consequences for the heart. This study had two purposes: first, to define the effects of ventricular fibrillation during hypoxia on energy metabolism and accumulation of intracellular Na' and, second, to test whether the occurrence of ventricular fibrillation can be predicted from functional or metabolic parameters. M...

Glucose and fatty acid metabolism in the isolated working mouse heart.

Although isolated perfused mouse heart models have been developed to study mechanical function, energy substrate metabolism has not been examined despite the expectation that the metabolic rate for a heart from a small mammal should be increased. Consequently, glucose utilization (glycolysis, oxidation) and fatty acid oxidation were measured in isolated working mouse hearts perfused with radiol...
