LncRNA gadd7, increased in varicocele patients, suppresses cell proliferation and promotes cell apoptosis

چکیده
Varicocele-related sperm damages are usually caused by oxidative stresses. Growing evidence indicates that lncRNA growth arrested DNA-damage inducible gene 7 (gadd7) is involved in the regulation of the oxidative stress responses. In this study, we measured the expression level of gadd7 in the sperm and found that the expression of gadd7 was significantly up-regulated in patients with varicocele compared with the healthy control. The relative expression level of gadd7 was negatively correlated with the sperm count. Overexpression of gadd7 suppressed cell proliferation and promoted cell apoptosis in mouse spermatocyte-derived cell lines GC-1 and GC-2. Furthermore, the protein level of Bax was raised while Bcl2 expression was reduced after overexpression of gadd7. This work provides a potential novel insight for the varicocele-related sperm impairment and male infertility.

LncRNA Miat Promotes Proliferation of Cervical Cancer Cells and Acts as an Anti-apoptotic Factor
There are a sub-population of cells in tumor tissues known as cancer stem cells (CSCs) which have similar features with stem cells, including self-renewal and differentiation capacity. Recently, it was established that not only stem cells factors such as Oct4, but also ES-associated lncRNAs are contributing to various regulatory aspects of CSCs. Myocardial infarction associated transcript (MIAT...

Background and Objective: Long non-coding ribonucleic acid (lncRNA) has been identified as an important gene regulator and prognostic marker in various cancers. The present study aimed to investigate the effects of Nuclear Paraspeckle Assembly Transcript1 (NEAT1) gene knockout using Clustered Regularly Interspaced Short Palindromic Repeats-associated Protein 9 (CRISPR/Cas9) in PC-3 cell line. ...
متن کاملKnockdown of lncRNA-PANDAR suppresses the proliferation, cell cycle and promotes apoptosis in thyroid cancer cells
Long non-coding RNAs (lncRNAs) have been found to show important regulatory roles in various human cancers. Lnc-RNA PANDAR is a novel identified lncRNA that was previously reported to show abnormal expression pattern in various cancers. However, little is known of its expression and biological function in thyroid cancer. Here, we used the quantitative real-time PCR (qRT-PCR) to determine the ex...

Background: MiR-103a-3p is a small non-coding RNA and has been reported to be involved in osteogenic proliferation and differentiation, but the role of miR-103a-3p in human osteoarthritis (OA) remains unclear. Objectives: In this study, we aimed to explore its function and molecular target in chondrocytes during OA pathogenesis. Materials an...
متن کاملDownregulated long non-coding RNA CLMAT3 promotes the proliferation of colorectal cancer cells by targeting regulators of the cell cycle pathway
Over-expression of long non-coding RNA (lncRNA)-CLMAT3 is significantly associated with colorectal liver metastasis and is an independent predictor of poor survival for colorectal cancer patients. However, as little is known regarding the role of this gene in the proliferation of colorectal cancer in vitro, we investigated the involvement of lncRNA-CLMAT3 in colorectal cancer cell proliferation...
