Dipeptidyl peptidase IV/CD26 in T cell activation, cytokine secretion and immunoglobulin production.

چکیده
Dipeptidyl peptidase IV (DPPIV/CD26; EC. 3.4.14.5), a widely distributed multifunctional type II plasma membrane glycoprotein, is involved in different biological processes. It is a serine protease associated with uptake and transmembrane transport of proline-containing peptides as well as with processing of physiological active peptides1. As an exopeptidase it cleaves N -terminal dipeptides after proline or alanine residues. Furthermore, an endopeptidase activity of DPPIV has also been reported2. Due to its interaction with proteins of the extracellular matrix (ECM), such as collagen and fibronectin, DPPIV/CD26 can also be considered as a cell adhesion molecule3. The interaction of DPPIV/CD26 with proteins of the ECM has been determined by several binding assays4-6. Although the binding properties of DPPIV/CD26 to proteins of the ECM have been well characterized, the significance of this interaction for its biological function, especially for the immune regulation is still unknown. It has been shown that DPPIV/CD26 plays a crucial role in T cell activation and immune regulation7-10. The expression level of DPPIV/CD26 is tightly regulated during the development of T lymphocytes and its density on the plasma membrane is markedly enhanced after lymphocyte activation.

Deficiency of CD26 results in a change of cytokine and immunoglobulin secretion after stimulation by pokeweed mitogen.
To investigate the role of CD26 in the immune system, CD26 gene knockout mice with C57BL/6 background were used to study the immune response after stimulation with PWM. CD26(-/-) mice display an apparently normal phenotype. However, in their spleen lymphocyte population the percentage of CD4(+) T cells is lower, and that of NK cells is higher, than that in CD26(+/+) mice. In their peripheral bl...
متن کاملActivity of Dipeptidyl Peptidase-IV/CD26 and Aminopeptidase N/CD13 in Secretome of Mesenchymal Stem Cells after Treatment with LPS and PMA
Background: Emerging evidence suggests that secretome of mesenchymal stem cells has many anti-inflammatory and regenerative properties, which makes it a suitable candidate for the treatment of autoimmune and degenerative diseases. Dipeptidyl Peptidase-IV (DPP-IV)/CD26 and Aminopeptidase N (APN)/CD13 are ubiquitous ecto-enzymes which can digest various substrates including some ...
متن کاملCD26-mediated signaling for T cell activation occurs in lipid rafts through its association with CD45RO.
CD26 is a T cell activation antigen that contains dipeptidyl peptidase IV activity and is known to bind adenosine deaminase. The mechanism by which CD26 costimulation potentiates T cell receptor-mediated T cell activation, leading to subsequent exertion of T cell effector function, is still not clearly defined. In this article, we demonstrate that CD26 localizes into lipid rafts, and targeting ...

The T cell marker CD26/dipeptidyl peptidase (DP) IV is associated with an effector phenotype and markedly elevated in the human CNS disorder multiple sclerosis. However, little is known about the in vivo role of CD26/DP IV in health and disease, and the underlying mechanism of its function in CNS inflammation. To directly address the role of CD26/DP IV in vivo, we examined Th1 immune responses ...
متن کاملGlycosaminoglycans Regulate CXCR3 Ligands at Distinct Levels: Protection against Processing by Dipeptidyl Peptidase IV/CD26 and Interference with Receptor Signaling
CXC chemokine ligand (CXCL)9, CXCL10 and CXCL11 direct chemotaxis of mainly T cells and NK cells through activation of their common CXC chemokine receptor (CXCR)3. They are inactivated upon NH₂-terminal cleavage by dipeptidyl peptidase IV/CD26. In the present study, we found that different glycosaminoglycans (GAGs) protect the CXCR3 ligands against proteolytic processing by CD26 without directl...
