چکیده

Although rhodanese activity occurs in many tissues (l-3), the enzyme had been obtained in crystalline form from beef liver only (4). Beef kidney rhodanese could not be crystallized by the same procedure (5). To extend the biochemical genetic comparison reported previously (5), however, it was desired to obtain the kidney enzyme in a very high state of purity. This paper reports the purification and crystallization of beef kidney rhodanese by a method which combines an ion exchange fractionation with steps based on Siirbo’s procedure for the crystalline liver enzyme (4). The same method has also been used for preparing crystalline beef liver rhodanese. Some further comparative data are presented.

Mechanism of Rhodanese Catalysis of Thiosulfate-lipoate Oxidation-reduction.

Crystalline beef liver rhodanese (thiosulfate : cyanidesulfurtransferase, EC 2.8.1.1) catalyzes sulfur transfer from SSOato CNby a double displacement mechanism (3, 4). The biological role of this reaction is not clear, but its possible involvement in CNdetoxication has been repeatedly stressed. The present study of the specificity of the crystalline beef liver enzyme and of the occurrence of t...

Rhodanese-catalyzed reduction of thiosulfate by reduced lipoic acid.

by a double displacement mechanism (1, 2). Thiosulfate and organic thiosulfonates will act as sulfur donors for the enzyme (3). Cyanide ion, SOS=, and the organic sulfinates will accept sulfur from the enzyme-sulfur compound (1, 4, 5). In this study, crystalline beef liver rhodanese has been found to catalyze the reduction of SSOp to HSand SOf. Several powerful reducing agents, such as NaBH4 an...

A comparison of some biochemical properties of the beef liver and beef kidney rhodaneses.

Several reports have indicated that there may be differences among the proteins responsible for a given enzymatic activity in difIerent tissues of the same animal (14). This paper reports the results of a comparative study of some biochemical properties of the enzyme rhodanese purified from the liver and the kidneys of the same steer. The two enyzmes have been found to be identical by all of th...

Mechanism of rhodanese action: isotopic tracer studies.

In a previous publication (2) a double displacement mechanism for the catalytic activity of rhodanese (thiosulfate cyanide sulfurtransferase) was proposed on the basis of polarographic evidence. In addition it was concluded from the polarographic data that crystahine beef liver rhodanese is an enzyme-substrate intermediate containing two activated substrate sulfur atoms per molecule. This commu...

The isolation of rhodanese from Pseudomonas aeruginosa by affinity chromatography.

Rhodanese (thiosulphate : cyanide sulphur-transferase; EC 2 . 8 . I . I) was originally ,described by Lang (1933). The enzyme catalyses the formation of thiocyanate from cyanide and thiosulphate according to the reaction : S20!+ CN-+ SCN+ SO:-. Rhodanese activity has been demonstrated in most mammalian tissues, with the greatest activity present in liver and kidney. The enzyme has been purified...
