Shigella Vaccine Development: The Model Matters
نویسنده
چکیده
Bacillary dysentery or shigellosis is mediated by the pathogenic enterobacteria Shigella. Shigella are transmitted through the fecal-oral route, no animal reservoir has been described, and remain a major cause of moderate-severe diarrhea worldwide since no Shigella vaccine is yet commercialized. The design of a pan-Shigella vaccine has been made difficult by the large variety of Shigella species and serotypes and the continuous identification of new serotypes due to the genomic plasticity of Shigella. The assessment of Shigella vaccine candidates remains complicated by the lack of a suitable animal model of shigellosis. Until know, several model of shigellosis have been characterized in the rhesus macaque, the rabbit, the mouse or the guinea pig, although none of them is satisfactory to assess the whole Shigella virulence process from an oral inoculation to the colonic mucosa colonization and disruption. The potential and limits of the different animal model of shigellosis used so far to study Shigella virulence mechanisms and to evaluate vaccine candidates’ efficacy are described here. Since no optimal animal model of shigellosis has been characterized, further efforts should be made to develop a suitable one, as a pre-requisite for the development of a successful Shigella vaccine.

Applying Mathematical Tools to Accelerate Vaccine Development: Modeling Shigella Immune Dynamics
We establish a mathematical framework for studying immune interactions with Shigella, a bacteria that kills over one million people worldwide every year. The long-term goal of this novel approach is to inform Shigella vaccine design by elucidating which immune components and bacterial targets are crucial for establishing Shigella immunity. Our delay differential equation model focuses on antibo...

Diarrheal diseases still remain health problem worldwide and out of many bacteria responsible for, Shigella and pathogenic Escherichia cause the most diarrheas in the world. Shigellacause bacterial dysenteries and shigellosis through invasion where the most effective proteins for pathogenesis is Ipac. Critical virulence protein for ETEC infection is CFA/I with two subunits called cfab and cfae....
متن کاملλ-Red-Recombineering Live Attenuated ΔipaD Shigella dysenteriae from Iranian Isolates as A Candidate of Vaccine
Shigella species (spp.) are gram-negative bacteria that are responsible for shigellosis. Although it can be controlled via antibiotics, increasing number of antibiotic resistant isolates of Shigella have been reported. Therefore, other strategies such as production of specific vaccine against this organism could be a suitable therapeutic approach. Attenuated live vaccines produced by gene delet...
متن کاملMonoclonal Antibodies to Shigella Lipopolysaccharide Are Useful for Vaccine Production.
There is a significant need for an effective multivalent Shigella vaccine that targets the most prevalent serotypes. Most Shigella vaccines under development utilize serotype-specific lipopolysaccharides (LPSs) as a major component based on protection and epidemiological data. As vaccine formulations advance from monovalent to multivalent, assays and reagents need to be developed to accurately ...

Background: Recombinant vaccine technology is one of the most developed means in controlling infectious diseases. However, an effective vaccine against Shigella is still missing. Objective: To evaluate recombinant IpaC protein of Shigella as a vaccine candidate. Methods: In this study we cloned IpaC gene into an expression vector in prokaryotic system. The protein expression was evaluated by SD...
