Cellular effects of gastric electrical stimulation on antral smooth muscle cells in rats.

چکیده
The cellular effects of gastric electrical stimulation (GES), which has recently been introduced as a potential therapy for the treatment of gastroparesis and obesity, were investigated in rat antrum smooth muscle cells (SMCs). Effects on cell membrane potentials of single electrical current pulses (pulse width from 0.1 ms to 200 ms) and 2-s pulse train stimuli with different pulse widths (0.1-4 ms), different frequencies (20-200 Hz), and different intensities were studied: 1) the stimulus amplitude had an exponential relationship to the pulse width from 2 ms to 200 ms, along with a rapidly rising strength-duration curve at pulse widths less than 5 ms, and a relatively flat curve at pulse widths greater than 50 ms; 2) when the pulse frequency was at 80 Hz or above, pulse train electrical stimulation, with a pulse width of 2 ms or above but not < or = 1 ms, was able to depolarize cell membrane potentials to above -30 mV and/or generate action potentials. Electrical stimulation with a single long pulse and a width of 50 ms or greater is effective in depolarizing cell membrane potentials of SMCs with low amplitude. Pulse train electrical stimulation with a pulse width of < or = 1 ms fails to generate action potentials in SMCs, whereas pulse train electrical stimulation with a pulse width of 2-4 ms and a sufficiently high pulse frequency is able to generate action potentials. These cellular findings may be useful in optimizing stimulation parameters of GES.

Enhancement of antral contractions and vagal afferent signaling with synchronized electrical stimulation.
Gastric filling activates vagal afferents involved in peripheral signaling to the central nervous system (CNS) for food intake. It is not known whether these afferents linearly encode increasing contractions of the antrum during antral distension (AD). The aim of this study was to investigate effects of AD and electrically enhanced antral contractions on responses of vagal afferents innervating...

Effects of acupuncture treatment on mechanical responses produced by transmural nerve stimulation (TNS) and acetylcholine (ACh) were investigated in circular smooth muscle preparations isolated from the antrum of the stomach of genetically hyperglycemic rats. While control rats had blood glucose levels of about 140 mg/dl, this was approximately tripled in the genetically hyperglycemic rats, but...

Sphingolipid metabolism was examined in guinea-pig airway smooth muscle cells stimulated by platelet-derived growth factor (PDGF) and 4β-phorbol 12- myristate 13-acetate (PMA), as mitogens and bradykinin (BK) as non-mitogen. Stimulation of the cells by PMA and PDGF for 60 min. at 37°C induced the following changes in sphingolipid metabolites: in cells prelabeled with PH] palmitate, a 1.2 f...
متن کاملEffects of Dopamine on the Contractility of the Circular Muscles in the Guinea-pig Gastric Antrum and Fundus'
=Abstract=The effects of dopamine on gastric motility were investigated using circular muscle strips prepared from the antral and' fundic regions of the guinea-pig stomachs. The spontaneous and phasic contractions induced by electrical stimulation were recorded with a force transducer. Dopamine potentiated the spontaneous contraction of the antral circular muscle, which was suppressed completel...
متن کاملInhibitory responses mediated by vagal nerve stimulation are diminished in stomachs of mice with reduced intramuscular interstitial cells of Cajal
Intramuscular interstitial cells of Cajal (ICC-IM) are closely associated with enteric motor nerve terminals and electrically coupled to smooth muscle cells within the gastric musculature. Previous studies investigating the role of ICC-IM in motor neurotransmission have used indiscriminate electric field stimulation of neural elements within the gastric wall. To determine the role of ICC-IM in ...
