چکیده
Background Of the 230,000 new cases of invasive breast cancer diagnosed annually, 70% express the estrogen receptor-a and may be treated with anti-estrogen therapies. While these drugs induce clinical benefit, over 50% will develop resistance. Recently, we have determined a key role of the unfolded protein response endoplasmic reticulum protein chaperone, glucose-regulated protein 78 (GRP78), in mediating anti-estrogen resistance. We now expand the role of GRP78 to include regulation of lipid metabolism.

An Evolutionary Relationship Between Stearoyl-CoA Desaturase (SCD) Protein Sequences Involved in Fatty Acid Metabolism
Background: Stearoyl-CoA desaturase (SCD) is a key enzyme that converts saturated fatty acids (SFAs) to monounsaturated fatty acids (MUFAs) in fat biosynthesis. Despite being crucial for interpreting SCDs’ roles across species, the evolutionary relationship of SCD proteins across species has yet to be elucidated. This study aims to present this evolutionary relationship based on amino aci...

The lipid composition of skin is important to a variety of functions served by this organ. Therefore, skin expresses multiple enzymes that synthesize and metabolize lipids. An important class of lipid metabolism enzymes expressed in skin is the lipid desaturases. Various isoforms of stearoyl-CoA desaturase, a delta-9 desaturase, as well as a delta-6 desaturase alter the lipid composition of the...

Stearoyl-CoA desaturase (SCD)1 converts saturated fatty acids into monounsaturated fatty acids. Using muscle overexpression, we sought to determine the role of SCD1 expression in glucose and lipid metabolism and its effects on exercise capacity in mice. Wild-type C57Bl/6 (WT) and SCD1 muscle transgenic (SCD1-Tg) mice were generated, and expression of the SCD1 transgene was restricted to skeleta...
متن کاملBiochemical and physiological function of stearoyl-CoA desaturase.
A key and highly regulated enzyme that is required for the biosynthesis of monounsaturated fatty acids is stearoyl-CoA desaturase (SCD), which catalyzes the D(9)-cis desaturation of a range of fatty acyl-CoA substrates. The preferred substrates are palmitoyl- and stearoyl-CoA, which are converted into palmitoleoyl- and oleoyl-CoA respectively. Oleate is the most abundant monounsaturated fatty a...
متن کاملEffects of glucose metabolism on the regulation of genes of fatty acid synthesis and triglyceride secretion in the liver.
Glucose disposal induces a signal that modulates the transcriptional regulation of genes involved in the glycolysis and lipogenesis pathways. To investigate the role of glucose metabolism on hepatic gene expression independently from insulin action, we overexpressed glucokinase, the limiting enzyme in the glycolysis pathway, in the liver of streptozotocin-induced type 1 diabetic rats. By microa...
