VASCULAR BIOLOGY NFATc3 regulates the transcription of genes involved in T-cell activation and angiogenesis

چکیده
The nuclear factor of activated T cells (NFAT) family of transcription factors plays important roles in many biologic processes, including the development and function of the immune and vascular systems. Cells usually express more than one NFAT member, raising the question of whether NFATs play overlapping roles or if each member has selective functions. Using mRNA knock-down, we show that NFATc3 is specifically required for IL2 and cyclooxygenase-2 (COX2) gene expression in transformed and primary T cells and for T-cell proliferation. We also show that NFATc3 regulates COX2 in endothelial cells, where it is required for COX2, dependent migration and angiogenesis in vivo. These results indicate that individual NFAT members mediate specific functions through the differential regulation of the transcription of target genes. These effects, observed on shortterm suppression by mRNA knock-down, are likely to have been masked by compensatory effects in gene-knockout studies. (Blood. 2011;118(3):795-803)

Angiogenesis, as the process of new vessel formation from pre-existing vessels is dependent on a delicate equilibrium between endogenous angiogenic and antiangiogenic factors. However, under pathological conditions, this tight regulation becomes lost which can result in the formation of the different diseases such as cancer. Angiogenesis is a complex process that includes many gene products tha...
متن کاملDiagnosis and Treatment B non-Hodgkin Lymphoma with System Biology Approaches
Lymphomas are solid tumors of immune system and Non-Hodgkin Lymphomas (NHL) is the most prevalent lymphomas; with wide ranges of histological and clinical features, it is so difficult to identify them. Herein, various bioinformatics tools (such as gene differential expressions, epigenetics and protein analysis) employed to find new treatment approach for NHL based on gene expression variation b...
متن کاملHuman activated macrophages and hypoxia: a comprehensive review of the literature
Macrophages accumulate in poorly vascularised and hypoxic sites including solid tumours, wounds and sites of infection and inflammation where they can be exposed to low levels of oxygen for long periods. Up to date, different studies have shown that a number of transcription factors are activated by hypoxia which in turn activate a broad array of mitogenic, pro-invasive, pro-angiogenic, and pro...
متن کاملTranscriptional Coactivator CBP Facilitates Transcription Initiation and Reinitiation of HTLV-I and Cyclin D2 Promoter
HTLV-I is the etiologic agent for adult T-cell leukemia/lymphoma (ATL) and HTLV-I-associated myelopathy/tropical spastic paraparesis (HAM/TSP). Taxi, the major activator of this virus, is a 40- kDa (353 amino acid) phosphoprotein, predominantly localized in the nucleus of the host cell, which functions to trans-activate both viral and cellular promoters. Recently it has been shown that HTLV-I a...
