چکیده

Ethidium bromide (EB) is a commonly used probe for fluorescence detection and quantification of nucleic acids, since EB forms a highly luminescent complex with dsDNA. Typical detection sensitivity of EB-based assays to dsDNA in solution is about 0.1 lg. With the aim to increase the sensitivity of EB assays we have employed the Metal-Enhanced Fluorescence technology. We show that deposition of the EB/DNA complex solutions onto the silver nanoparticle surface additionally increases the far-field observable fluorescence 5-fold, yielding a net fluorescence enhancement factor of 180-fold, as compared to the free dye in solution. Subsequently, our approach enables the more sensitive detection of dsDNA. Published by Elsevier B.V.

Direct observation of the reversible unwinding of a single DNA molecule caused by the intercalation of ethidium bromide

Ethidium bromide (EtBr) is the conventional intercalator for visualizing DNA. Previous studies suggested that EtBr lengthens and unwinds double-stranded DNA (dsDNA). However, no one has observed the unwinding of a single dsDNA molecule during intercalation. We developed a simple method to observe the twisting motions of a single dsDNA molecule under an optical microscope. A short dsDNA was atta...

Fluorescence Lifetime Spectroscopy in Mu1tipleScattenng Environments: An Application to Biotechnology

Over the past few years, there has been significant research activity devoted to the application of fluorescence spectroscopy to strongly scattering media, where photons propagate diffusely. Much ofthis activity focused on fluorescence as a source of contrast enhancement in optical tomography. Our efforts have emphasized the quantitative recovery of fluorescence parameters for spectroscopy. Usi...

The fluorescence resonance energy transfer (FRET) gate: a time-resolved study.

The two-step energy-transfer process in a self-assembled complex comprising a cationic conjugated polymer (CCP) and a dsDNA is investigated by using pump-dump-emission spectroscopy and time-correlated single-photon counting; energy is transferred from the CCP to an ethidium bromide (EB) molecule intercalated into the dsDNA through a fluorescein molecule linked to one terminus of the DNA. Time-d...

DNA Interaction Studies of Selected Polyamine Conjugates

The interaction of polyamine conjugates with DNA double helix has been studied. Binding properties were examined by ethidium bromide (EtBr) displacement and DNA unwinding/topoisomerase I/II (Topo I/II) activity assays, as well as dsDNA thermal stability studies and circular dichroism spectroscopy. Genotoxicity of the compounds was estimated by a comet assay. It has been shown that only compound...
