چکیده

DEDICATION To my family ii ACKNOWLEDGEMENTS As I come to the completion of one chapter of my life and prepare to begin the next, I reflect upon a quote I have carried with me throughout my graduate studies from the African-American educator and author Booker T. Washington-" Success is to be measured not so much by the position that one has reached in life as by the obstacles by which s(he) has overcome while trying to succeed ". As I prepare to complete my PhD and leave The University of Michigan after six years, I reflect on the journey that brought me here, the experiences that shaped my time as a PhD student, and most importantly the people who were there along the way. I have been fortunate to have many people guiding and supporting me, and I am FOREVER indebted to them all. The first two people I want to thank are my advisors, Dr. Helen A. Baghdoyan and Dr. Ralph Lydic. Their role in my graduate education as both mentors and teachers was invaluable and I am grateful to them for their dedication to me throughout this journey. I would also like to thank the other members of my doctoral committee, Dr. whose guidance and feedback during my graduate years was much appreciated. I cannot forget to thank the faculty, staff, and students of both the Pharmacology and Anesthesiology Departments for all of their support and help along the way, many of you will remain lifelong friends. iii Last, but certainly not least, I would like to thank ALL of my family and friends who have been with me throughout this journey, especially my parents and my sister. Your love, encouragement, and support made so much of this dream a reality. Words cannot express what you all mean to me, but know that my success is your success.

GABA(A) receptors in the pontine reticular formation of C57BL/6J mouse modulate neurochemical, electrographic, and behavioral phenotypes of wakefulness.

Drugs that potentiate transmission at GABA(A) receptors are widely used to enhance sleep and to cause general anesthesia. The mechanisms underlying these effects are unknown. This study tested the hypothesis that GABA(A) receptors in the pontine reticular nucleus, oral part (PnO) of mouse modulate five phenotypes of arousal: sleep and wakefulness, cortical electroencephalogram (EEG) activity, a...

Immediate Epileptogenesis after Kainate-Induced Status Epilepticus in C57BL/6J Mice: Evidence from Long Term Continuous Video-EEG Telemetry

The C57BL/6J mouse as a model of seizure/epilepsy is challenging due to high mortality and huge variability in response to kainate. We have recently demonstrated that repeated administration of a low dose of kainate by intraperitoneal route can induce severe status epilepticus (SE) with 94% survival rate. In the present study, based on continuous video-EEG recording for 4-18 weeks from epidural...

Ganaxolone suppression of behavioral and electrographic seizures in the mouse amygdala kindling model.

Ganaxolone (3alpha-hydroxy-3beta-methyl-5alpha-pregnan-20-one), a synthetic analog of the endogenous neurosteroid allopregnanolone and a positive allosteric modulator of GABAA receptors, may represent a new treatment approach for epilepsy. Here we demonstrate that pretreatment with ganaxolone (1.25-20 mg/kg, s.c.) causes a dose-dependent suppression of behavioral and electrographic seizures in ...

Neuroactive steroids for the treatment of status epilepticus.

Benzodiazepines are the current first-line standard-of-care treatment for status epilepticus but fail to terminate seizures in about one third of cases. Synaptic GABAA receptors, which mediate phasic inhibition in central circuits, are the molecular target of benzodiazepines. As status epilepticus progresses, these receptors are internalized and become functionally inactivated, conferring benzo...

Spontaneous unitary discharges of the mesencephalic reticular formation during sleep and wakefulness in normal and chronically blinded cats.

An acute blockade of the retinal tonic discharges in midpontine pretrigeminal cats is found effective to synchronize the cortical EEG which otherwise maintains the desynchronized pattern1, 2. It is suggested that the tonic retinal discharges energize the ascending reticular activating system to maintain the cerebral cortex in the desynchronized state. To test the validity of this surmise in the...
