Analysis of the promoter activity of a wheat dehydrin gene (DHN-5) under various stress conditions

چکیده
A wheat dehydrin gene (DHN-5) is inducible by either treatment with abscisic acid (ABA) or by abiotic stress such as drought and salinity. To further investigate the regulation of the gene, a 1,128-bp genomic fragment upstream of the DHN-5 translated sequence has been isolated, cloned, and designated as the ‘‘PrDHN-5’’ promoter. Sequence analysis of PrDHN-5 revealed the presence of cisregulatory elements which could be required for abiotic stress and ABA responsiveness. The PrDHN-5 promoter was fused to the glucuronidase (gusA) gene and the resulting construct was transferred into Arabidopsis thaliana (heterologous dicotyledonous system) and wheat (homologous monocot system). Histochemical assays of stably transformed Arabidopsis plants showed that PrDHN-5 is active in this heterologous system. The accumulation of gusA transcripts was up-regulated by salt, osmotic and ABA treatment. Histochemical assay in wheat embryos showed a staining in callus and younger leaves treated with various stress. Altogether, these results show that PrDHN-5 is an abiotic-stress-inducible promoter.

Role of the durum wheat dehydrin in the function of proteases conferring salinity tolerance in Arabidopsis thaliana transgenic lines.
Dehydrins are claimed to stabilize macromolecules against freezing damage, dehydration, ionic or osmotic stresses, thermal stress and re-folding yield. However, their precise function remains unknown. In this context, we report the behavior of protease activities in dehydrin transgenic Arabidopsis lines against the wild type plant under salt stress (100mM NaCl). Indeed, proteases play key roles...
متن کاملThe K-segments of the wheat dehydrin DHN-5 are essential for the protection of lactate dehydrogenase and β-glucosidase activities in vitro.
The wheat dehydrin DHN-5 has been previously shown to exhibit heat protecting effect on enzymatic activities. In order to understand the molecular mechanism by which DHN-5 exerts its protective function, we performed an approach to dissect the functional domains of DHN-5 responsible for this feature. In two distinct enzymatic assays, we found that the truncated forms of DHN-5 containing only on...

Drought is the cause of adverse environmental impacts on plant growth and crop yield. In this study, the effects of water deficit on plant molecular and physiological responses were investigated using two cultivars (namely, Sardari and Zarin) of bread wheat selected based on the results of a three-year research. For the purposes of this study, they were grown in plastic pots containing field so...

The effect of drought stress on the relative expression of Aquaporin, Dehydrin, Putative leafy cotyledon 1-like protein (Ha-L1L) genes in two sunflower lines [ENSAT254 (tolerant) and LC1064C (susceptible)] was investigated using real time PCR technique. The lines with different response to drought stress were planted using completely randomized design with three replications in greenhouse of fa...
متن کاملThe K-segments of wheat dehydrin WZY2 are essential for its protective functions under temperature stress
Dehydrins (DHNs), group 2 of late embryogenesis abundant (LEA) proteins, are up-regulated in most plants during cold, drought, heat, or salinity stress. All DHNs contain at least one K-segment, which is believed to play a significant role in DHN function by forming an amphipathic helix. In previous studies, wzy2, an YSK2-type DHN gene, was isolated from the Zhengyin 1 cultivar of Triticum aesti...
