چکیده

Angiotensin II (AII) evokes a biphasic increase in inositol 1,4,5-trisphosphate (Ins(1,4,5)P3) levels in adrenal glomerulosa cells, with an extracellular Ca(2+)-independent early peak followed by a secondary sustained elevation that is highly dependent on the presence of extracellular Ca2+. The Ca(2+)-dependent sustained phase of agonist-induced Ins(1,4,5)P3 production was closely correlated with Ca2+ influx and was inhibited by inorganic Ca2+ channel blockers with the potency ratio: La3+ >> Cd2+ > Mn2+ > Co2+ > Ni2+. Of the two Ca2+ surrogates, Sr2+ and Ba2+, Sr2+ was partially active compared with Ca2+, and Ba2+ was inactive in restoring Ins(1,4,5)P3 formation in cells stimulated with AII in Ca(2+)-free medium. However, unlike Ca2+, Sr2+ only weakly supported and Ba2+ failed to affect the calmodulin-activation of Ins(1,4,5)P3 3-kinase. Also, there was an accumulation of Ins(1,4,5)P3 and diminished formation of Ins(1,3,4,5)P4 and Ins(1,3,4)P3 when intact glomerulosa cells were stimulated by AII in the presence of Sr2+. This difference between the Sr2+ sensitivity of phospholipase C and Ins(1,4,5)P3 3-kinase provides a means for the potentiation of agonist-induced elevations of Ins(1,4,5)P3 in the intact cell and for direct analysis of the role of the inositol tris-/tetrakisphosphate pathway in cellular signaling.

Multiple pathways of inositol polyphosphate metabolism in angiotensin-stimulated adrenal glomerulosa cells.

The production and metabolism of inositol 1,4,5-trisphosphate (Ins-1,4,5-P3) and other inositol polyphosphates was studied in cultured bovine adrenal glomerulosa cells prelabeled for 24 h with [3H]inositol. During stimulation with angiotensin II, Ins-1,4,5-P3 increased to a peak of 15-fold above basal within 10 s, followed by a second phase of continuous increase over the next 30 min. Ins-1,4,5...

Angiotensin-stimulated production of inositol trisphosphate isomers and rapid metabolism through inositol 4-monophosphate in adrenal glomerulosa cells.

The production and metabolism of inositol phosphates in rat adrenal glomerulosa cells prelabeled with [3H]inositol and stimulated with angiotensin II were analyzed by high-performance anion-exchange chromatography. Exposure to angiotensin II was accompanied by a rapid and substantial decrease in the phospholipid precursor, phosphatidylinositol (PtdIns) 4,5-bisphosphate with only a slight and tr...

Inositol 1,3,4,5-tetrakisphosphate stimulates calcium release from bovine adrenal microsomes by a mechanism independent of the inositol 1,4,5-trisphosphate receptor.

In bovine adrenal microsomes, Ins(1,4,5)P3 binds to a specific high-affinity receptor site (Kd = 11 nM) with low affinity for two other InsP3 isomers, Ins(1,3,4)P3 and Ins(2,4,5)P3. In the same subcellular fractions Ins(1,4,5)P3 was also the most potent stimulus of Ca2+ release of all the inositol phosphates tested. Of the many inositol phosphates recently identified in angiotensin-II-stimulate...

Source of 3H-labeled inositol bis- and monophosphates in agonist-activated rat parotid acinar cells.

The kinetics of [3H]inositol phosphate metabolism in agonist-activated rat parotid acinar cells were characterized in order to determine the sources of [3H]inositol monophosphates and [3H]inositol bisphosphates. The turnover rates of D-myo-inositol 1,4,5-trisphosphate and its metabolites, D-myo-inositol 1,4-bisphosphate and D-myo-inositol 1,3,4-trisphosphate, were examined following the additio...

Inositol 1,4,5-trisphosphate receptor subtypes in adrenal glomerulosa cells.

Expression of the diverse subtypes of inositol 1,4,5-trisphosphate (InsP3) receptor (IP3R) was examined in rat adrenal glomerulosa cells. The polymerase chain reaction products were characterized by means of DNA sequencing and/or restriction enzyme mapping. The predominant subtype expressed is IP3R-1; its alternatively spliced variants containing and lacking segment S1 are present in comparable...
