چکیده

The diagnostic value of circulating SEPT9 DNA methylation in patients with colorectal cancer remains controversial. Thus, we performed a systematic review and meta-analysis to assess diagnostic accuracy of SEPT9 DNA methylation for colorectal cancer (CRC). We conducted a comprehensive literature search in PubMed, the Cochrane library, Web of Science databases and Chinese National Knowledge Infrastructure (CNKI) up until January 20, 2016. The diagnostic sensitivity (SEN), specificity (SPE), positive likelihood ratio (PLR), negative likelihood ratio (NLR), diagnostic odds ratio (DOR), and summary receiver operating characteristic (SROC) curve were pooled using STATA 12.0 software. A total of 6,079 subjects included 1,890 colorectal cancer patients in 20 studies were included in this meta-analysis. The summary estimates for circulating methylated SEPT9 DNA in the diagnosis of CRC in these studies were pooled SEN =0.72 (95% confidence interval [CI]: 0.65-0.78), SPE =0.88 (95% CI: 0.850.91), PLR 6.13 (95% CI: 4.90-7.67), NLR 0.32 (95% CI: 0.25-0.40]), and DOR 19.3 (95% CI: 13.14-28.31), and the area under the curve (AUC) was 0.90 (95% CI: 0.87-0.92). The current analysis indicated that circulating methylated SEPT9 DNA may be a valuable marker in the diagnosis of CRC, but this conclusion has to be interpreted cautiously owing to high heterogeneity and some limitations. Further studies with more well-designed should be required to confirm the results in the future.

Circulating methylated SEPT9 DNA in plasma is a biomarker for colorectal cancer.

BACKGROUND The presence of aberrantly methylated SEPT9 DNA in plasma is highly correlated with the occurrence of colorectal cancer. We report the development of a new SEPT9 biomarker assay and its validation in case-control studies. The development of such a minimally invasive blood-based test may help to reduce the current gap in screening coverage. METHODS A new SEPT9 DNA methylation assay ...

Diagnostic Performance of DNA Hypermethylation Markers in Peripheral Blood for the Detection of Colorectal Cancer: A Meta-Analysis and Systematic Review

DNA hypermethylation in blood is becoming an attractive candidate marker for colorectal cancer (CRC) detection. To assess the diagnostic accuracy of blood hypermethylation markers for CRC in different clinical settings, we conducted a meta-analysis of published reports. Of 485 publications obtained in the initial literature search, 39 studies were included in the meta-analysis. Hypermethylation...

Detection of aberrant methylated SEPT9 and NTRK3 genes in sporadic colorectal cancer patients as a potential diagnostic biomarker.

Colorectal cancer (CRC) is one of the most common malignancies, and the third leading cause of cancer mortality worldwide. Timely detection of CRC in patients with earlier stages provides the highest rate of survival. Epigenetic alterations are important in the occurrence and progression of CRC, and represent the primary modifications of cancer cells. Therefore, detection of these alterations i...

Septin 9 promoter region methylation in free circulating DNA—potential role in noninvasive diagnosis of lung cancer: preliminary report

Currently, there are no sensitive diagnostic tests that could allow early detection of lung cancer. Among some cancer patients, epigenetic changes in the nature of methylation of different gene promoter regions are observed, which affect expression of suppressor genes such as septin 9 (SEPT9). Due to the ability of detecting these changes in free circulating DNA in peripheral blood, such genes ...

Detection of Methylated Septin 9 in Tissue and Plasma of Colorectal Patients with Neoplasia and the Relationship to the Amount of Circulating Cell-Free DNA

BACKGROUND Determination of methylated Septin 9 (mSEPT9) in plasma has been shown to be a sensitive and specific biomarker for colorectal cancer (CRC). However, the relationship between methylated DNA in plasma and colon tissue of the same subjects has not been reported. METHODS Plasma and matching biopsy samples were collected from 24 patients with no evidence of disease (NED), 26 patients w...
