چکیده

Photoactivation allows one to pulse-label molecules and obtain quantitative data about their behavior. We have devised a new modeling-based analysis for photoactivatable actin experiments that simultaneously measures properties of monomeric and filamentous actin in a three dimensional cellular environment. We use this method to determine differences in the dynamic behavior of beta and gamma actin isoforms, showing that both inhabit filaments that depolymerize at equal rates but that beta actin exists in a higher monomer-to-filament ratio. We also demonstrate that Cofilin equally accelerates depolymerization of filaments made from both isoforms, but is only required to maintain the beta actin monomer pool. Finally, we used modelling-based analysis to assess actin dynamics in axon-like projections of differentiating neuroblastoma cells, showing that the actin monomer concentration is significantly depleted as the axon develops. Importantly, these results would not have been obtained using traditional half time analysis. Given that parameters of the publicly available modeling platform can be adjusted to suit the user’s own experimental system, this method can easily be used to quantify actin dynamics in many different cell types and subcellular compartments. Simultaneous measurements of actin filament turnover, filament fraction, and monomer diffusion in endothelial cells.

The analogous techniques of photoactivation of fluorescence (PAF) and fluorescence recovery after photobleaching (FRAP) have been applied previously to the study of actin dynamics in living cells. Traditionally, separate experiments estimate the mobility of actin monomer or the lifetime of actin filaments. A mathematical description of the dynamics of the actin cytoskeleton, however, predicts t...

Measuring Actin Dynamics

The actin cytoskeleton is a highly dynamic system responsible for maintaining cell shape and rigidity. In endothelial cells, the actin cytoskeleton undergoes a remarkable reorganization in response to shear at the cell surface. This rearrangement likely occurs through regulation of cytoskeletal dynamics. This thesis presents a viable technology for the measurement of two fundamental dynamic pro...

Enhanced Depolymerization of Actin Filaments by ADF/Cofilin and Monomer Funneling by Capping Protein Cooperate to Accelerate Barbed-End Growth

A living cell's ability to assemble actin filaments in intracellular motile processes is directly dependent on the availability of polymerizable actin monomers, which feed polarized filament growth [1, 2]. Continued generation of the monomer pool by filament disassembly is therefore crucial. Disassemblers like actin depolymerizing factor (ADF)/cofilin and filament cappers like capping protein (...

A theoretical analysis of filament length fluctuations in actin and other polymers.

Control of the structure and dynamics of the actin cytoskeleton is essential for cell motility and for maintaining the structural integrity of cells. Central to understanding the control of these features is an understanding of the dynamics of actin filaments, first as isolated filaments, then as integrated networks, and finally as networks containing higher-order structures such as bundles, st...
