چکیده

Areas of preneoplastic livers characterized by intense RNA staining apparently represent the sites of neoplastic transformation. These hyperbasophilic foci, as well as the hepatomas, showed a marked decrease in the stain ing intensity of the cytoplasmic proteins in sections stained by the mercury-bromophenol blue method or the MillÃ3nreaction. Following extraction of the RNA, how ever, protein stainability in these sites was comparable to that of surrounding parenchyma. Radioautographic studies further indicated that hyperbasophilic foci and hepatomas do not differ from the surrounding tissue in the labeling of cytoplasmic proteins by tritiated leucine. These results were interpreted as indicating the pres ence in sites of neoplastic transformation and liver tumors of some unusual RNA which binds to and masks the cytoplasmic proteins.

Changes in polyamine levels and protein synthesis rate during rat liver carcinogenesis induced by 4-dimethylaminoazobenzene.

The concentrations of putrescine, spermidine, and spermine in liver of rats fed on 4-dimethylaminoazobenzene and in the resultant hepatomas were found to be significantly higher than were those observed in normal liver from rats of the same strain, sex, and age. These modifications were due to the carcinogen and not to the special low-riboflavin diet used to obtain the carcinogenic effect of 4-...

Spectrophotometric analysis of cytochromes in rat liver during carcinogenesis.

The cytochrome content of liver mitochondria and microsomes during the carcinogenesis of rat liver by both 3'-methyl-4-dimethylaminoazobenzene (3'-DAB) and 4'methyl-4-dimethylaminoazobenzene (4'-DAB), and of 3'DAB-induced hepatomas, was determined spectrophotometrically at room and liquid nitrogen temperatures and was compared with that of normal liver. The cyto chrome a + a3 content in the liv...

Alteration in the distribution of basic soluble rat liver proteins during azo dye carcinogenesis.

Antibodies against rat liver // proteins and against a similar charge class of azoproteins have been labeled with fluorescein isothiocyanate (FITC), and the labeled antisera have been used as histoserological stains on unfixed frozen sections of rat liver. Fixation of antibody was defined by fluorescence microscopy and was used to determine the intracellular distribution of the appropriate anti...

Fine Structural Alterations in Cell Particles During Chemical Carcinogenesis

The previous studies on the correlation between tumor incidence and changes in microsomal and mitochondrial swelling during feeding of 3'-methyl-4-dimethylaminoazobenzene to rats have been extended to other hepatic carcinogens. Administration of 4'-fluoro-4-dimethylaminoazobenzene, 4'-ethyl-2-methyl-4-dimethylaminoazobenzene, 2-acetylam-inofluorene, ethionine, and tannic acid were found to prod...

The histochemical localization of alkaline phosphatase during carcinogenesis in rats fed p-dimethylaminoazobenzene.

Greenstein in 1942 found a high content of alka line phosphatase in transplanted rat hepatoma 31 (3). This tumor arose originally in the liver of a male Osborne-Mendel rat and was induced by the basal diet of White and Jackson (16) containing 0.06 percent />-diinethylaniinoazobenzene (DAB) with intermittent addition of 0.5 per cent cystine and 0.5 per cent methionine (17). The enzyme ac tivity ...
