چکیده

In this, the first of three papers, we present the sequence of the ribosomal RNA (rRNA) genes of Drosophila melanogaster. The gene regions of D. melanogaster rDNA encode four individual rRNAs: 18S (1,995 nt), 5.8S (123 nt), 2S (30 nt), and 28S (3,945 nt). The ribosomal DNA (rDNA) repeat of D. melanogaster is AT rich (65.9% overall), with the spacers being particularly AT rich. Analysis of DNA simplicity reveals that, in contrast to the intergenic spacer (IGS) and the external transcribed spacer (ETS), most of the rRNA gene regions have been refractory to the action of slippage-like events, with the exception of the 28S rRNA gene expansion segments. It would seem that the 28S rRNA can accommodate the products of slippage-like events without loss of activity. In the following two papers we analyze the effects of sequence divergence on the evolution of (1) the 28S gene "expansion segments" and (2) the 28S and 18S rRNA secondary structures among eukaryotic species, respectively. Our detailed analyses reveal, in addition to unequal crossing-over, (1) the involvement of slippage and biased mutation in the evolution of the rDNA multigene family and (2) the molecular coevolution of both expansion segments and the nucleotides involved with compensatory changes required to maintain secondary structures of RNA.

Evidence that intergenic spacer repeats of Drosophila melanogaster rRNA genes function as X-Y pairing sites in male meiosis, and a general model for achiasmatic pairing.

In Drosophila melanogaster males, X-Y meiotic chromosome pairing is mediated by the nucleolus organizers (NOs) which are located in the X heterochromatin (Xh) and near the Y centromere. Deficiencies for Xh disrupt X-Y meiotic pairing and cause high frequencies of X-Y nondisjunction. Insertion of cloned rRNA genes on an Xh- chromosome partially restores normal X-Y pairing and disjunction. To map...

Aconitase and Developmental EndPointsasEarly IndicatorsofCellularToxicity Induced by Xenobiotics in Drosophila Melanogaster

Background: In this study, the toxicity of the different xenobiotics was tested on the fruit fly Drosophila melanogaster model system.  Methods: Fly larvae were raised on food supplemented with xenobioticsat different concentrations (sodium nitroprusside (0.1-1.5 mM), S-nitrosoglutathione (0.5-4 mM), and potassium ferrocyanide (1 mM)). Emergence of flies, food intake by larvae, and pupation h...

Concentration dependent effect of morphine, aspirin, capsaicin and chili pepper hydro alcoholic extract on thermal and chemical pain model in fruit fly (Drosophila melanogaster)

Introduction: Pain research using animal models is related to ethical concerns, so invertebrates and insects have been recommended by researchers. In the present study, the nociceptive and antinociceptive effects of capsaicin, aspirin, morphine and chili extract were examined using fruit fly (Drosophila melanogaster) as an alternative for rodent pain model. Methods: Stage 3 of larvae and ad...

A comparative phylogenetic analysis of Theileria spp. by using two two "18S ribosomal RNA" and "Theileria annulata merozoite surface antigen" gene sequences

More than 185 species, strains and unclassified Theileria parasites are categorized in the Entrez Taxonomy. The accurate diagnosis and proper identification of the causative agents are important for understanding the epidemiology, prevention and appropriate treatment. This study aims to discuss the importance of two genes of Theileria annulata 18S ribosomal RNA (18S rRNA) and Theileria annulata...
