Genetic Analysis of the Induction of Tumors by Methylcholanthrene
نویسنده
چکیده
Reference has been made to the observation that mutations and other sudden bi'ologic changes occur in the untreated descendants of mice that in each of several generations have been given a single subcutaneous injection of methylcholanthrene.'0 It has been suggested that some of these changes can be explained by point mutations while others must have involved a greater biologic alteration. An analysis of one of these more widespread alterations is the basis for this communication.

RNA tumor virus gs antigen and tumor induction by various doses of 3-methylcholanthrene in various strains of mice treated as weanlings.
The effect of various doses of 3-methylcholanthrene on s.c. tumor induction and the occurrence of the murine C-type RNA group specific (gs) viral antigen in tumor tissue were studied in weanling mice of 8 genotypically different strains. Tumor incidence was found to be related to the dose of 3-methylcholanthrene. On the other hand, the incidence of gs antigen in the induced tumors was independe...
متن کاملDose response and growth rates of subcutaneous tumors induced with 3-methylcholanthrene in mice and timing of tumor origin.
The dose response of tumor induction after single s.c. injections of 3-methylcholanthrene (MC) into the groin of mice and the growth rates of the tumors formed were examined. The dose-response curve of tumor induction appeared to be linear at low doses of MC. Analysis of 182 sarcomas produced between 50 and 400 days after MC injection into WB and C3H/He mice, together with previous data on ICR/...
متن کاملAnalysis of Exon 19 and 39 of ATM Gene in Brain Tumors; Considering the P53 Accumulation
Many studies have been focused on cytogenetic and molecular genetic defects in brain tumors; therefore the role of ATM as a tumor suppressor gene in these tumors is poorly considered. In this study mutation analysis of exon 19 and 39 of ATM gene and P53 accumulation were investigated by PCR-SSCP, sequencing, and flow cytometry . Four polymorphisms including D1853N, IVS 38-8 T?C, F858L, P872T we...
متن کاملAcceleration of the Development of Leukemia and Induction of Tumors in Akr Mice by Neonatal Injection of 20-methylcholanthrene.
A single subcutaneous injection of 20-methylcholanthrene into newborn AKR mice less than 24 hours old resulted in the acceleration of the development of lymphocytic leukemia, and induction of subcutaneous sarcomas and multiple-lung adenomas. Morphological descriptions of the respective tumors were given. It is suggested that the lungs of newborn mice of strain AKR may prove to be a sensitive or...
متن کاملThe genetic basis of susceptibility to leukemia induction in mice by 3- methylcholanthrene applied percutaneously
Susceptibility to leukemia induction in mice by skin painting with 3-methylcholanthrene (MCA) is strain-specific, occurring only in strains relatively resistant to MCA-induced skin tumors. The Ah locus, which has a dominant allele (Ahb) for inducibility of the aryl hydrocarbon hydroxylase (AHH) enzyme system and a recessive allele (Ahd) for noninducibility, appears to be the major determinant o...
