چکیده

This chapter is focused on the development of experiments and theory of using solid-state nanopores for sensing single protein molecules in their native and unfolded states. Proteins serve diverse roles such as transport carriers, catalysts, molecular motors, cellular structural support, and others that make life possible. Because of these widely differing roles, proteins have an enormously diverse set of shapes, sizes, and charge structures as compared to polynucleic acids. Solid-state nanopores are particularly suitable for characterizing single proteinmolecules because they can be fabricated with adjustable dimensions and are stable under conditions that denature proteins. This chapter describes the nanopore experimental setup, signal recording, data analysis, and basic principles related to the experiments and the theory connecting the electrical signal with the properties of proteins. Examples of experimental results illustrate the ability of solid-state nanopores to differentiate proteins in their folded and unfolded states. Native-state protein nanopore translocation follows biased one-dimensional diffusion of charged particles that is sensitive to size and electrical charge. Due to the heterogeneous charge sequence of polypeptides, unfolded proteins obey a coupled electrophoretic and thermally activated process that is sequence specific. The chapter concludes with a discussion of future directions and open challenges for single protein characterization using solid-state nanopores.

Integration of solid-state nanopores in microfluidic networks via transfer printing of suspended membranes.

Solid-state nanopores have emerged as versatile single-molecule sensors for applications including DNA sequencing, protein unfolding, micro-RNA detection, label-free detection of single nucleotide polymorphisms, and mapping of DNA-binding proteins involved in homologous recombination. While machining nanopores in dielectric membranes provides nanometer-scale precision, the rigid silicon support...

Stochastic sensing of proteins with receptor-modified solid-state nanopores.

Solid-state nanopores are capable of the label-free analysis of single molecules. It is possible to add biochemical selectivity by anchoring a molecular receptor inside the nanopore, but it is difficult to maintain single-molecule sensitivity in these modified nanopores. Here, we show that metallized silicon nitride nanopores chemically modified with nitrilotriacetic acid receptors can be used ...

Multiplexed ionic current sensing with glass nanopores.

We report a method for simultaneous ionic current measurements of single molecules across up to 16 solid state nanopore channels. Each device, costing less than $20, contains 16 glass nanopores made by laser assisted capillary pulling. We demonstrate simultaneous multichannel detection of double stranded DNA and trapping of DNA origami nanostructures to form hybrid nanopores.

Engineering solid-state nanopores for detection of single transcription factors bound to DNA

Detection and characterization of nucleic acid-protein interactions, particularly those involving DNA and transcription factors, remain significant barriers to our understanding of genetic regulation. Solid-state nanopores are extremely sensitive single molecule sensors with the capability to map local chemical and structural characteristics along the length of a biopolymer, providing label-fre...
