چکیده

Dakin and Dudley (1) and Keuberg (2) discovered the enzyme glyoxalase in 1913, and showed that it could convert methylglyoxal to lactic acid. The usual assay for this enzyme is a manomctric procedure (3). In 1928 Ariyama (4) described a method for the calorimetric determination of glyoxals involving the ability of these compounds to reduce Benedict’s uric acid reagent (5) in an alkaline medium. Under his optimal conditions, Ariyama measured between 580 and 870 y of glyoxal. By halving the volumes of the reagents he could detect as little as 607 of glyoxal. A modification of Ariyama’s procedure, to be described herein, enables one to measure as little as 2 y of methylglyoxal and also permits the determination of glyoxalase activity of tissues by examining the disappearance of methylglyoxal.

Glyoxalase activities in human tumour cell lines in vitro.

The activities of glyoxalase I (EC 4.4.1.5) and glyoxalase II (EC 3.1.2.6) were measured in 3 human non-malignant and 28 tumour cell lines. The activity of glyoxalase I was in the range 321-8751 mUnits/mg of protein. The glyoxalase I/glyoxalase II activity ratio was in the range 7-147; glyoxalase I activity was always markedly greater than glyoxalase activity. There was no significant differenc...

Effect of Linalool on the Activity of Glyoxalase-I and Diverse Glycation Products in Rats with Type 2 Diabetes

Background and purpose: Hyperglycemia contributes to type 2 diabetes and diabetes vascular complications by reduction of the activity of glyoxalase-I (GLO-I) and elevation of glycation, oxidative stress, and inflammatory markers. Linalool is reported to have beneficial effects on glucose metabolism in animal models of diabetes, so, this study aimed at investigating the effect of linalool on the...

Stress-induced changes of methylglyoxal level and glyoxalase I activity in pumpkin seedlings and cDNA cloning of glyoxalase I gene

Abiotic stresses cause extensive losses to agricultural production worldwide. In this study, the effects of various abiotic stresses on the upregulation of methylglyoxal levels and glyoxalase I activities in pumpkin seedlings (Cucurbita maxima Duch.) were investigated. Most of the stresses caused significant increases in methylglyoxal level and glyoxalase I activity, white light causing the hig...

Dietary Fructose Feeding Increases Adipose Methylglyoxal Accumulation in Rats in Association with Low Expression and Activity of Glyoxalase-2

Methylglyoxal is a precursor to advanced glycation endproducts that may contribute to diabetes and its cardiovascular-related complications. Methylglyoxal is successively catabolized to D-lactate by glyoxalase-1 and glyoxalase-2. The objective of this study was to determine whether dietary fructose and green tea extract (GTE) differentially regulate methylglyoxal accumulation in liver and adipo...

Effect of antidiabetic compounds on glyoxalase I activity in experimental diabetic rat liver.

The activity of glyoxalase I from the soluble fraction of diabetic rat liver was found to decrease as compared to the control. Sodium orthovanadate in drinking water and Trigonella foenum graecum seed powder when administered to these diabetic animals were found to reverse the activity of glyoxalase I to control values. A combination of the above two antidiabetic compounds showed a better rever...
