Short communication Involvement of NMDA receptors in the antidepressant-like action of adenosine

چکیده
Background and Method: In this work, the contribution of NMDA receptors to the antidepressant-like effect of adenosine in the forced swimming test (FST) was investigated. Results: The pretreatment of mice with NMDA or D-serine was able to prevent the anti-immobility effect of either adenosine or MK-801 in the FST. In addition, the administration of a sub-effective dose of adenosine produced a synergistic effect with subeffective doses of MK-801, ketamine and zinc chloride. Moreover, the immobility time of the mice treated with active doses of adenosine or N6-cyclohexyladenosine (CHA) plus MK-801 was not significantly different from that obtained with adenosine, CHA and MK-801 alone; by contrast, the combination between active doses of adenosine and CHAplus an active dose of the tricyclic antidepressant imipramine produced a greater effect in the FST than the administration of either drug alone. Conclusion: Together, the results suggest that the effect of adenosine in the FST is likely dependent on the inhibition of NMDA receptors mediated by the activation of adenosine A1 receptors.

Short communication Involvement of NMDA and AMPA receptors in the antidepressant-like activity of antidepressant drugs in the forced swim test
Background: The involvement of glutamate system (particularly the NMDA and AMPA receptors) in the mechanism of antidepressant activity was demonstrated in preclinical and clinical studies. Methods: In the present study, we investigated the effect of NMDA and AMPA receptors’ ligands (agonists and antagonists) on the antidepressant-like activity of escitalopram, milnacipran, imipramine and reboxe...

متن کاملEffect of imipramine and desipramine on adenosine receptors in isolated rat atria
The effect of different doses (1-50 µ M) of imipramine (IMI) and desipramine (DES) on the rate and force of contraction of isolated rat atria was studied. IMI and DES produced a dose-dependent increase in force of contraction (31- 94% for IMI and 35-118% for DES). Pretreatment of rats with reserpine (5 mg/kg) on the isolated atria with propranolol (1 µ g) inhibited the positive ionotropic eff...

Objective(s): Studies indicate that major deficiency in the levels of monoaminergic transmitters is a reason for severe depression. On the other hand, it is shown that Carthamus tinctorius L. (CT) may improve neuropsychological injuries by regulation of the monoamine transporter action. Hence, the present study was undertaken to evaluate the involvement of monoaminergic systems in antidepressan...
