Therapeutic Discovery HDL Mimetics Inhibit Tumor Development in Both Induced and Spontaneous Mouse Models of Colon Cancer

چکیده
Recent studies suggest that high-density lipoprotein (HDL) levels are inversely related to colon cancer risk. HDL mimetics constructed from a number of peptides and proteins with varying structures possess anti-inflammatory and antioxidant properties reminiscent of HDL. In this article, we examined whether HDLmimetics, L-4F (an apolipoprotein A-I mimetic peptide) and G (an apolipoprotein J mimetic peptide) affect tumor growth and development in mouse models of colon cancer. HDL mimetics reduced viability and proliferation of CT26 cells, a mouse colon adenocarcinoma cell line, and decreased CT26 cell–mediated tumor burden in BALB/c mice when administered subcutaneously or orally. Plasma levels of lysophosphatidic acid (LPA), a serum biomarker for colon cancer, were significantly reduced in mice that received HDL mimetics, suggesting that binding and removal of proinflammatory lipids is a potential mechanism for the inhibition of tumor development by HDL mimetics. Furthermore, L-4F significantly reduced size and number of polyps in APCmin/þ mice, a mouse model for human familial adenomatous polyposis, suggesting that HDL mimetics are effective in inhibiting the development of both induced and spontaneous cancers of the colon. Our results, for the first time, identify HDL mimetics as a novel therapeutic strategy for the treatment of colon cancer. Mol Cancer Ther; 11(6); 1311–9. 2012 AACR.

متن کاملChanges in monocarboxylate transporter 1 and p53 gene expression by aerobic interval training in the experimental colon carcinoma of mouse
Background: Recent evidence suggests that regular exercise training is effective in treating various aspects of cancer. Therefore, the purpose of this study was to determine the effect of 8 weeks of aerobic interval training on monocarboxylate transporter 1 (MCT1) protein and expression of p53 gene in tumor of colon cancer mice. Methods: The present study was conducted experimentally from May ...

Despite diagnostic, preventive and therapeutic advances, growing incidence of cancer and high rate of mortality among patients affected by specific cancer types indicate current clinical measures are not ideally useful in eradicating cancer. Chemoresistance and subsequent disease relapse are believed to be mainly driven by the cell-molecular heterogeneity of human tumors that necessitates perso...

متن کاملAnticancer Properties of Chrysin on Colon Cancer Cells, In vitro and In vivo with Modulation of Caspase-3, -9, Bax and Sall4
Background: The SALL4/Sall4 is constitutively expressed in human and mice. SALL4 mRNA could be used as a marker for the diagnosis of different types of cancers. On the other hand, chrysin has diverse biological properties. Objectives: In the present study, the effect of the chrysin was investigated on the CT26 colon cancer in vitro and in vivo. Furthermore, the expression levels of the stem cel...
