derivation of es-like cell from neonatal mouse testis cells in autologous sertoli cells co-culture system

چکیده
background: objective: this study presents an economical and simple co-culture system for pluripotent stem cells generation from neonatal mouse testis materials and methods: isolated testicular cells were cultured in dmem/f12. characteristics of the isolated cells and obtained es-like cell were immune-cytochemically confirmed by examining the presence of plzf, vimentin, oct4 and nanog protein. expression of the pluripotency and germ-cell specific genes was analyzed by qpcr in derived es-like colony and sscs respectively. results: the experiment results indicated that our method of obtaining pluripotent es-like cells from spermatogonial cells (scs) is simpler than the described methods. es-like cells were immunopositive for pluripotency markers. es-like cell qpcr results indicated significant increase in pluripotency genes expression and significant decrease in germ cell-specific genes expression. conclusion: the results indicated that es-like cell with pluripotency characteristic were generated from freshly isolated spermatogonial cells. the pluripotent stem cells provide a cellular reservoir usable for regenerative medicine instead of embryonic stem cells. spermatogonial stem cell (ssc) is a self-renewing population of male adult stem cell. sscs have a differentiation potential which are similar to embryonic stem cells. these embryonic stem like (es-like) cells can be a potential source for pluripotent cells for stem cell-based therapy.

Derivation of ES-like cell from neonatal mouse testis cells in autologous sertoli cells co-culture system
BACKGROUND Spermatogonial stem cell (SSC) is a self-renewing population of male adult stem cell. SSCs have a differentiation potential which are similar to embryonic stem cells. These Embryonic stem like (ES-like) cells can be a potential source for pluripotent cells for stem cell-based therapy. OBJECTIVE This study presents an economical and simple co-culture system for pluripotent stem cell...

objective: this study presents a simple method for isolation, expansion and purification of neonatal mouse spermatogonial stem cells. methods: we used enzymatic digestion to isolate a cell suspension of spermatogonia and sertoli cells from neonatal 2-day-old mice. the cells were cultured in dmem/f12 that contained 10% serum for two weeks. sertoli and spermatogonia cell characteristics were conf...
متن کاملCo-culture of Mouse Embryonic Stem Cells with Sertoli Cells Promote in vitro Generation of Germ Cells
Objective(s): Sertoli cells support in vivo germ cell production; but, its exact mechanism has not been well understood. The present study was designed to analyze the effect of Sertoli cells in differentiation of mouse embryonic stem cells (mESCs) to germ cells. Materials and Methods: A fusion construct composed of a Stra8 gene promoter and the coding region of enhanced green fluorescence p...
متن کاملa simple co-culture system for generation of embryonic stem-like cells from testis
background new research proposes the pluripotency of spermatogonial cells obtained from testis. these spermatogonia-derived stem cells are called embryonic stem-like cells that express embryonic stem cell markers and differentiate to the three germ layers. objectives the aim of the present study was to generate embryonic stem-like cells from neonatal mouse testis. materials and methods the test...
متن کاملSertoli cell condition medium can induce germ like cells from bone marrow derived mesenchymal stem cells
Objective(s): Although many researchers have confirmed induction of germ cells from bone marrow mesenchymal stem cells (BMMSCs), there are no reports that confirm spontaneous differentiation of germ cells from BMMSCs. In this study, we have evaluated the effect of adult Sertoli cell condition medium (SCCM) as a mutative factor in the induction of germ cells from BMMSCs. Materials and Methods: ...
